Rat Cartilage Repair Using Nanophase PLGA/HA Composite and Mesenchymal Stem Cells

Abstract: Biodegradable polymer/bioceramic composite poly(lactide- coglycolide)/hydroxyapatite(PLGA/HA) was studied for bone tissue engineering. The PLGA/HA composite was evaluated as a scaffold with the ability for mesenchymal stem cells (MSC) to participate in cartilage repair. The PLGA/HA composite and the PLGA/HA composite cultured with MSC were transplanted into cartilage defects created in rats. The PLGA/HA-MSC and PLGA/HA had better tissue morphology, structure integrity, matrix staining, and much thicker newly f… Show more

“…The pre-clinical data with regards to allogenic cells is conflicting. One group showed promising results of allogenic MSCs in a rabbit model when compared to autologous cells, although numbers were small [ 160 , 161 ]. Another group compared autologous chondroprogenitor cells and allogenic chondroprogenitor cells against controls in an equine model and reported inferior repair in the allogenic cell group [ 23 ].…”

“…But, due to the deficiency of the natural strength of molecular recognition sites on the PLGA side, they can bound the PLGA application in the medical area to a specific point and inhibit the specific cell adhesion (Chen et al, 2006;Hsu et al, 2006;Zhang et al, 2016). Therefore, materials with higher cell affinity, for example, hydroxyapatite and collagen, are mostly reformed with PLGA for the enhancement of biocompatibility (Kawazoe et al, 2009;Li, Zheng, et al, 2009). Wool keratin is a natural biomaterial (Tachibana et al, 2002(Tachibana et al, , 2005Yamauchi et al, 1996;Zhang et al, 2016).…”
