Ratiometric Fluorescent Safranin-O Staining Allows the Quantification of Lignin Contents In Muro

Abstract: In some specific vascular plant tissues, lignin can impregnate the entire cell wall to make it more rigid and hydrophobic. Different techniques have been developed in the past years to make possible the quantification of this polyphenolic polymer at the organ or tissue level but difficulties of access to the cellular level remain. Here we describe an approach based on ratiometric emission measurements using safranin-O and the development of a macro adapted for the FIJI software which makes possible the quantif… Show more

“…The authors of [51][52][53] administered exogenous paclobutrazol to winter wheat cultivars that are tolerant/vulnerable to lodging and discovered that paclobutrazol dramatically decreased the volume of the wheat internode, promoted lateral growth, and increased the accumulation of lignin, the operation of the lignin biosynthesis enzyme, and thickness of the internode, thus increasing the resistance to wheat lodging. Lignin has the ability to permeate the entire cell wall in the tissues of some species of vascular plant, increasing its rigidity and hydrophobicity [54]. According to a different study, crop density has a considerable impact on the morphological properties of the stem and lignin biosynthesis, thereby enhancing the stem's mechanical power and lowering the impact of lodging [55].…”
