Mung bean (Vigna radiata L. Wilczek) categorized as one of the pivotal annual crops of Vigna genera is commonly cultivated in rotation with the cereal crops during the drought season. Conversely, to ameliorate its stunted productivity, the interspecific hybridization technique has been introduced between the mung bean and the common bean to promote genetic improvement with the breeding projects in Indonesia. However, since mung bean is a self-pollinated crop and has a narrow genetic base, the selection and improvement of a specific trait using marker-assisted selection is more challenging. Hence, a precautionary investigation is imperative to evaluate the progenies resulting from interspecific hybridization using an ideal marker. This study aimed to investigate the genetic variability of the F2 population of the interspecific mung bean hybrids using retrotransposon-based markers, particularly Inter-Retrotransposon Amplified Polymorphism (IRAP) markers. In this study, we identified retrotransposon from the mung bean genome and determined the Long Terminal Repeat (LTR) sequence using the LTR Finder. The IRAP primers were designed from a conserved region of the LTR sequence. One hundred of the F2 interspecific hybrids generated from the crossing between mung bean and common bean were successfully discriminated by IRAP markers. The IRAP marker showed high heterozygosity and moderate Polymorphic Information Content (PIC) values. The IRAP markers were able to detect genetic variability in the F2 progenies resulting from the interspecific hybridization. Cluster analysis showed that 100 of the F2 progenies were grouped into three clusters. This study demonstrated that retrotransposon-based markers can offer an effective approach for evaluating the segregation in the F2 population of intercross hybrids in the mung bean.

Background: Despite the best efforts for improving the greengram varieties, the yield potential of this crop remains low owing to both biotic and abiotic factors. Yield losses caused due to various diseases were estimated to be 24-67 per cent in anthracnose, 80-100 per cent in mungbean yellow mosaic virus and 40 per cent due to powdery mildew So, the use of resistant cultivars is the most effective and eco-friendly method of managing the disease. Methods: A series of field experiments spanned over seven years (2014-2021) were conducted at Main Agricultural Research Station, University of Agricultural Sciences, Dharwad, Karnataka. In the first set of experiments, the material comprised of eight varieties of greengram which were treated with 20kR gamma radiation. M1was planted in Kharif 2014 and each variety was bulk harvested. M2 was planted in summer 2014 and individual plant selection was carried out. Mutant families were advanced to the further generation in Kharif 2015 and Rabi 2015. The M2 mutants were scored for their reaction to anthracnose, cercospora leaf spot and powdery mildew diseases with respective susceptible and resistant checks. In another set of experiments aimed at developing MYMV tolerant genotypes, an investigation was carried out with 168 F2 individuals derived from crossing Vigna trilobata and DGGV2. One row infector line of DGGV-2 was raised after every three test entries to study the reaction to MYMV. Three rows of susceptible check was planted all around the border to ensure enough inoculum load. Of the 168 F2 individuals, two lines showed resistance to MYMV. They were stabilized through a series of selfing generations. Result: Evaluation of advanced breeding lines for their reaction to various foliar diseases revealed that the advanced breeding lines DGG-203, DGG-206, DGG-208, DGG-21, DGG-80, DGG-273, DGG-274, DGG-275, DGG-262, DGG-263, DGG-264, DGG-82, DGG-200, TARM-1 (C), DGG-215-5, DGG-302, DGG-152, IPM-3-2, DGG-20, DGG-73, IPM-14-10, DGG-121, DGG-250, DGG-109, DGG-96 had seed yields ranging from 9.8 g to 12.4 g and moderately resistant to both anthracnose and CLS. The breeding lines DGG-203, DGG-206, DGG-21, DGG-273, IPM-3-2 (Check), DGG-96 have recorded combined resistance to anthracnose, cercospora leaf spot and powdery mildew diseases.

Kersting's groundnut [Macrotyloma geocarpum (Harms.) Maréchal and Baudet], Fabaceae, is an important source of protein and essential amino acids. As a grain legume species, it also contributes to improving soil fertility through symbiotic nitrogen fixation. However, the crop is characterized by a relatively low yield (≤500 kg/ha), and limited progress has been made so far, toward the development of high-yielding cultivars that can enhance and sustain its productivity. Recently, there was an increased interest in alleviating the burdens related to Kersting's groundnut (KG) cultivation through the development of improved varieties. Preliminary investigations assembled germplasms from various producing countries. In-depth ethnobotanical studies and insightful investigation on the reproductive biology of the species were undertaken alongside morphological, biochemical, and molecular characterizations. Those studies revealed a narrow genetic base for KG. In addition, the self-pollinating nature of its flowers prevents cross-hybridization and represents a major barrier limiting the broadening of the genetic basis. Therefore, the development of a research pipeline to address the bottlenecks specific to KG is a prerequisite for the successful expansion of the crop. In this paper, we offer an overview of the current state of research on KG and pinpoint the knowledge gaps; we defined and discussed the main steps of breeding for KG' cultivars development; this included (i) developing an integrated genebank, inclusive germplasm, and seed system management; (ii) assessing end-users preferences and possibility for industrial exploitation of the crop; (iii) identifying biotic and abiotic stressors and the genetic control of responsive traits to those factors; (iv) overcoming the cross-pollination challenges in KG to propel the development of hybrids; (v) developing new approaches to create variability and setting adequate cultivars and breeding approaches; (vi) karyotyping and draft genome analysis to accelerate cultivars development and increase genetic gains; and (vii) evaluating the adaptability and stability of cultivars across various ecological regions.
