Refined localization of the Escherichia coli F4ab/F4ac receptor locus on pig chromosome 13

Abstract: Diarrhoea in newborn and weaned pigs caused by enterotoxigenic Escherichia coli (ETEC) expressing F4 fimbriae leads to considerable losses in pig production. In this study, we refined the mapping of the receptor locus for ETEC F4ab/F4ac adhesion (F4bcR) by joint analysis of Nordic and Swiss data. A total of 236 pigs from a Nordic experimental herd, 331 pigs from a Swiss experimental herd and 143 pigs from the Swiss performing station were used for linkage analysis. Genotyping data of six known microsatellite m… Show more

“…The positions of the 18 SNPs are consistent with previously reported QTL regions [3], [9]. These results offer a high possibility that F4abR and F4acR share the common causal mutation(s) as suggested by some previous studies [5], [7], [29].…”

“…Once the bacteria adhere to the brush border of enterocytes and colonize the small intestine, the enterotoxins produced by them induce an increased secretion of electrolytes into lumen, resulting in diarrhoea. However, not all piglets are susceptible to ETEC F4 and the susceptibility is determined by the existence of a specific receptor of ETEC F4ab/F4ac [3].…”

“…The location of F4ab/acR was then refined to a region between Sw207 and S0075 on SSC13q41 in this population (Jørgensen et al 2003). More recently, F4ab/acR was narrowed to the SW207-S0075 interval on SSC13 and the most likely position was in the SW207-MUC4 region (Joller et al 2009). Those encoding mucin-like sialoglycoproteins and transferrin-like proteins (Erickson et al 1994;Grange and Mouricout 1996) in the refined region are most likely positional candidate genes.…”

“…Moreover, several candidate genes (Joller et al, 2006;Peng et al, 2007;Zhang et al, 2008) for the F4ab/ac and F18 receptors (Meijerink et al, 2000) have been proposed. a These authors contributed equally to this study.…”
