Regulation of Interleukin 2 Receptor Expression by Interleukin 2

Abstract: The regulatory influence of Interleukin 2 (IL-2) on the expression of IL-2 receptors (IL-2R) was studied using long-term cultured T-cell lines and recombinant IL-2 (r-IL-2). Three T-cell lines with different growth requirements were used as model systems: insulin-specific BK-BI-1.2 cells express IL-2R transiently after antigenic restimulation, ovalbumin-reactive BK-OVA-1R cells express IL-2R permanently, and BK-BI-2.6.C6 cells bear IL-2R constitutively but do not exhibit antigen reactivity. All three T-cell li… Show more

“…The IL-2R expression by Leu 3+ antigen-induced T-cell lines from PB of normal donors reported here is in full accord with the results of others (4,16,17). IL-2R expression in all studies reached a maximum within 4 days after mitogenic or antigenic stimulation and decreased thereafter despite the presence of saturating IL-2 concentrations.…”

“…were cultured with OVA at 100 pug/ml in Iscove's modified Dulbecco's medium (IMDM; GIBCO), supplemented with 2 mM L-glutamine, 50 ,uM 2-mercaptoethanol, 100 international units of penicillin, streptomycin at 100 ,zg/ml, and 5% heat-inactivated fetal calf serum (Seromed, Berlin) (complete medium). After BK-OVA-1R T cells with specificity for OVA were established as described (19). Clone ST2/K.9 T cells (20) are reactive to bovine insulin (BI, a generous gift of G. Seipke, Hoechst, Frankfurt).…”

“…Clone ST2/K.9 T cells (20) are reactive to bovine insulin (BI, a generous gift of G. Seipke, Hoechst, Frankfurt). BK-BI-2.6.04.1 is an Ia-positive sister clone of BK-BI-2.6.C6 (6). These three lines are of (B10 x B10.BR)F1 origin and express interleukin 2 (IL-2) receptors constitutively.…”

“…BK-BI-1.2 T cells, described in [lo], recognize the glutamic acid residue in position 4 of the A chain of BI in the context of I-Abxk restriction molecules. BK-BI-7 T cells, established as reported in [13], are specific for amino acids A8-AlO within the A chain loop of BI, seen in combination with I-Abxk Ia molecules. BK-OVA-1: establishment and characteristics of this T cell line with specificity for ovalbumin (OVA) were reported before [ 131.…”
