Regulation of T lymphocyte subpopulations in specific pathogen-free chickens following experimental fowl adenovirus-Ⅷinfection

Abstract: Two-day-old specific pathogen-free (SPF) chickens were divided into two groups. Group I was inoculated orally with fowl adenovirus Ⅷ (FAV-Ⅷ). Group II served as a negative control. Chickens were investigated at various days post-inoculation (dpi) by flow cytometric analysis for changes in T lymphocyte subpopulations in immune system and blood. In the thymus, CD3+T lymphocytes were increased at 25 dpi, with significant increases in the FAV infected noted at 1, 12, 20dpi (p<0.05). This was accompanied by a corre… Show more

“…Live-attenuated vaccines mimic natural infections and should induce cellular immunity [ 49 ]. High levels of CD3 T cells have been reported among chickens inoculated with FAdV 8 up to 25 dpi [ 50 ], but CD3+, CD4+, and CD8+ cells were induced up to 42 dpi in this trial. These findings were more remarkable in the liver, which was the primary organ of infection at day 35 pi, but also in the spleen and thymus at different time points.…”

“…The thymus, where T cells mature, plays an important role in the development of cellular immunity [ 50 ]. However, FAdV 4 reduced the percentage of CD4+ and CD8+ cells in the thymus [ 13 ].…”

“…The undulation in the CD3+ T-cell sub-population in the inoculated chickens is a sign of T-lymphocyte inducement, an essential requirement for overcoming viral infections [51]. Montanide 71VG induced high cellular immunity when coupled with inactivated avian influenza virus vaccine in chicken [52] and could have aided the inducement of CMI in this trial. The CD3+ protein complex was first identified by immunoprecipitation experiments using human T-cells [53] and is a defining feature of the T-cell lineage; therefore, anti-CD3 antibodies can be used effectively as T-cell markers [53].…”
