Relative sensitivity of fish and mammalian cells to the antibiotic, trimethoprim: cytotoxic and genotoxic responses as determined by neutral red retention, Comet and micronucleus assays

Abstract: Relative cytotoxicity and genotoxicity of a widely used antibiotic, trimethoprim (TRIMP) was evaluated under in vitro conditions using rainbow trout gonad-2 (RTG-2) and Chinese hamster ovary-K1 (CHO-K1) cells. Whilst cytotoxicity was determined using neutral red retention (NRR) assay, the genotoxicity was determined using single cell gel electrophoresis or the Comet assay and cytokinesis-block micronucleus (CBMN) assay. For NRR assay, concentration-dependent cytotoxic effect was observed for both the cell line… Show more

“…These results therefore confirm the sensitivity of fish cells to genotoxic compounds and they also show the necessity to include different endpoints to address the genotoxic potential of a mixture or a chemical. In some cases, fish cells have been shown to be more sensitive than mammalian cells as regards the induction of genetic damage (Papis et al, 2011).…”

“…induction of chromosomal aberrations, micronuclei), DNA repair and cell-cycle measurements (Ng et al 2010). These end points apply to both human-related toxicity in vitro (human and mammalian cells), as well for ecogenotoxicity evaluation (Dixon et al 2002; Jha 2004; Raisuddin and Jha 2004; Jha 2008; Papis et al 2011). For MNMs it is worth keeping this broad perspective in testing strategies so that relative risk to both wildlife and humans can be assessed.…”

“…Papis et al, 60 evaluated the in vitro cytotoxic and genotoxic effects of trimethoprim on fish and mammalian cell by neutral red retention, comet and micronucleus assays. Concentration-dependent cytotoxic effect of trimethoprim was observed for both the cell lines by all the three tests.…”
