Review on Role of Natural Αlpha-Glucosidase Inhibitors for Management of Diabetes Mellitus

Abstract: Diabetes, a state of improperly regulated homeostasis of carbohydrate and lipid metabolism is one of the major killers in recent times. Most prevalent form of diabetes is non insulin dependent diabetes mellitus (NIDDM/type II). Rapid hydrolysis of starch mediated by pancreatic α-amylase and α-glucosidases followed by glucose uptake at intestine results in sudden rise in blood glucose levels, causing hyperglycemia in type II diabetes patients. Alpha-glucosidase inhibitors act as competitive inhibitors of enzyme… Show more

“…Rhazya stricta is claimed to contain various medicinal properties including antihyperglycemic effects like some other members of Apocynaceae family e.g., Telosma procumbens Merr., Vinca rosea L., Picralima nitida [ 47 ] . Root extracts of the plant inhibited α-glucosidase enzyme that can lead to better glycemic control through a reduction in disaccharide hydrolysis [ 48 ]. The inhibition of the DPP-IV enzyme is another strategy to keep incretin hormones available in the blood for insulin-dependent disposal of glucose from the blood.…”

“…Currently, the screening of the inhibitors of α-amylase and α-glucosidase of natural origin has received attention because this would avoid the side effects of commercial inhibitors for treating type-2 diabetes [ 10 ]. Extracts of natural plants with inhibitory α-glucosidase activity, such as tea and raspberry, are recommended as substitutes for synthetic drugs [ 11 ].…”

“…Carbohydrate hydrolyzing enzymes including α-glucosidase may be utilized for blocking the breakdown of starch and disaccharide into glucose and made glucose less available for absorption into the blood and ultimately sugar elevated level into the blood can be controlled. 5 Protein tyrosine phosphatase 1B (PTP1B) is considered as the main regulator of body stored body fat, insulin resistance, and energy balance. Insulin controls various functions at the level of gene transcription, protein translation, and enzyme activity.…”
