“…PI-MEC can function as alveolar progenitors in subsequent pregnancies, and it is thought that they would be related to differences in response to hormonal stimulation and carcinogenic agents observed between nulliparous and parous females (Wagner et al 2002, Henry et al 2004, Boulanger et al 2005. Several authors have focused on finding molecular changes as a mechanism of the pregnancy-induced protection (Russo & Russo 1994, Ginger et al 2001, Sivaraman et al 2001, D'Cruz et al 2002, Ginger & Rosen 2003, Medina & Kittrell 2003, Medina 2004. These changes in the genomic signature induced by physiological process like pregnancy have significant relevance with the novel observation from our studies relating that prenatal exposure to low and high doses of BPA induced differences in the gene expression of the mammary glands, which were time-and dose specific.…”

“…This fact may explain the low mean values of hCG-mRNA copy numbers in our study (mean of breast carcinomas: 68 copies per 200 ng total RNA; mean of fibroadenomas: 85 copies). Russo & Russo (1994) showed that hCG was able to induce a protective effect on rat mammary carcinogenesis and that the effect was dose dependent. After carcinogen administration, rats treated with hCG developed both benign and malignant tumours, but their incidence and the number of tumours per animal were significantly decreased by hormonal treatment.…”

“…This finding is not surprising because the two stages have diametrically opposed cell fates and are structurally, hormonally, and functionally distinct. The developing mammary gland is primarily composed of ductal epithelium proliferating under the influence of estrogen and progesterone, whereas involuting mammary gland is lobulo-alveolar epithelium undergoing apoptosis initiated by prolactin withdrawal (37,38). The marked differences at these stages likely account for the disparate effects of IGF-II on PTEN.…”
