Role of Reduced Sarco-Endoplasmic Reticulum Ca2+-ATPase Function on Sarcoplasmic Reticulum Ca2+ Alternans in the Intact Rabbit Heart

Abstract: Sarcoplasmic reticulum (SR) Ca2+ cycling is tightly regulated by ryanodine receptor (RyR) Ca2+ release and sarco-endoplasmic reticulum Ca2+-ATPase (SERCA) Ca2+ uptake during each excitation–contraction coupling cycle. We previously showed that RyR refractoriness plays a key role in the onset of SR Ca2+ alternans in the intact rabbit heart, which contributes to arrhythmogenic action potential duration (APD) alternans. Recent studies have also implicated impaired SERCA function, a key feature of heart failure, i… Show more

“…Furthermore, optical mapping enables direct contactless recording of optical action potentials, whereas multi electrode array techniques make indirect recordings of extracellular field potentials which require direct or close electrode-tissue contact 8 , 9 . By using calcium indicators, optical mapping can also be used to directly image calcium transients, the rise and fall of intracellular calcium concentration that initiates the contraction and relaxation of cardiac muscle cells 10 , 11 . These features make optical mapping an extremely powerful pre-clinical technique for studying arrhythmogenesis and the electrophysiological effects of physiological, pathophysiological, and pharmacological stimuli.…”

“…To unravel these unexcepted findings, we next undertook a pharmacological approach to address the ability of the TKO mice to resist hypothermia. Treatment of control and TKO mice with the selective SERCA inhibitor cyclopiazonic acid (CPA) (Wang, Myles et al 2021) reduced core body temperature at room temperature (Figure 2A). As expected, CPA treatment resulted in a faster rate of decline and to a lower extent when the mice were exposure to 4 o C (Figure 2B).…”

“…Therefore, unsurprisingly, attenuation of IR-induced alterations in calcium-handling protein expression could not reflect its functional roles during TH in this study. Furthermore, both hypothermia-induced RYR2 and SERCA2a dysfunction promote the emergence of SDA ( Wang et al, 2021 ). Because rotigaptide has no significant effects on Ca i homeostasis, the modulation of Ca i dynamics, for example, direct or indirect CaMKII inhibition ( Mitsuyama et al, 2014 ; Chou et al, 2020a ), may work synergistically with rotigaptide to suppress arrhythmogenic alternans in our model.…”
