Selection of Vaccinia Virus-Neutralizing Antibody from a Phage-Display Human-Antibody Library

Abstract: of ~0.16 mg/ml (~1.07 µM). This VC212 could serve as a valuable starting material for further development of VACV-neutralizing human immunoglobulin for a prophylactic measure against post-vaccination complications and for post-exposure treatment against smallpox.

“…Conventional methods of screening mice or rabbits for neutralizing antibodies might be too slow for this outbreak. Still, rapid methods (such as phage or yeast display libraries that express antibody fragments) can be used to identify virally neutralizing antibody candidates quickly [59,60]. The simplest, most direct way to combat SARS-CoV-2 during the outbreak is to interfere with the viral entry into cells, where antibodies usually work [61].…”

“…As per distributed research of the genome grouping of COVID-19 (GenBank: MN908947.3), specialists can deliver quality union in the lab and consider the S protein as an immunogen. For killing antibodies, conventional methods of screening mice or hares could be utilized that might be unreasonably delayed for this flare-up (Keck et al 2019;Shin et al 2019). Every immune response possibility to affirm killing COVID-19 and forestall contamination is important tried in cell culture and creature models and that has become a test.…”

“…[71][72][73][74] Moreover, another strategy proposed for COVID-19 treatment is targeting the SARS-CoV-2 surface S protein using neutralizing antibody (nAbs). 75 Besides, targeting the SARS-CoV-2 viral RNA genome by the use of RNA interference (RNAi) or antisense oligonucleotides is known as another interesting approach to have a potential therapy of this disease. 76,77 In Nano-based drug delivery systems, several evidences indicated that multiple nanomaterials such as graphene, nanodiamonds, carbon nanotubes, and polystyrene particles, have an intrinsic capacity to act as anti COVID-19 agents, especially via activating the immune system, which mostly depends on their functionalization.…”
