“…The cultures of V. tessellata have been established following our previous work (Manokari et al, 2020). The hardening and acclimatization steps determine the successful development of micropropagation protocol for V. tessellata and considered as a maturation phase.…”

“…The mature and healthy pods (2–4 cm long) of Vanda tessellata were collected from the Auroville Botanical Garden, Puducherry, India. The cultures were established and plantlets were produced by following the recent report of Manokari, Latha, Priyadharshini, Phanikanth, and Shekhawat (2020) on in vitro propagation of V. tessellata . Briefly, the protocorms were developed from seeds on full strength Murashige and Skoog's (MS) medium (Murashige & Skoog, 1962), containing 2.0 mg L −1 6‐benzylaminopurine (BAP), and somatic embryogenic potential of protocorms was stimulated using MS medium containing 1.0 mg L −1 BAP and 0.5 mg L −1 indole‐3 acetic acid (IAA).…”

“…Hence, there is a need to consider the type of explants used. Potential explants include the use of seeds (Utami & Hariyanto, 2019) as previously studied by Adhikari & Pant (2019), Dulić et al (2019), Godo et al (2020), and Manokari et al (2021). However, this study differs from previous studies, we provided C. rochussenii orchid seeds taken directly from the wildlife reserve area of Bukit Rimbang and Baling wildlife reserve areas.…”

“…Increasing the length of rewarming time had no significant effect on survival, suggesting that the most critical events occur within the first minute of rewarming. Manokari et al (2021) established an efficient protocol for asymbiotic seed germination of the grey orchid (Vanda tessellata (Roxb.) Hook.…”
