Simultaneous Quantitation and Validation of Triterpenoids and Phytosteroids in Phaseolus angularis Seeds

Abstract: A reproducible analytical method using reverse-phase high liquid performance chromatography combined with UV detecting was developed for the quantitative determination of four compounds isolated from the ethanol extract of Phaseolus angularis seeds (PASE): oleanolic acid (1), oleanolic acid acetate (2), stigmasterol (3) and β-sitosterol (4). This method was fully validated in terms of linearity (r 2 > 0.999), accuracy (98.5%-100.8%), precision (<0.92%), LOD (<0.0035 mg/mL), and LOQ (<0.0115 mg/mL). The effects… Show more

“…Mostly, HPLC detection coupled with different extraction methods and detection sources has been used for the analysis of oleanolic and ursolic acids in medicinal herbs and plants. [5][6][7][8][9][10] Some studies describe the application of gas 3,4 and liquid chromatography 11 coupled with a mass spectrometric detector, high-performance thin-layer chromatography (HPTLC), 1,12 supercritical uid chromatography with an evaporative light scattering detector (SFC-ELSD) 13 and a reversed phase-ultra ow liquid chromatography (RP-UFLC) 14 method for the determination of these triterpenoic acids.…”

“…4B, the isolated compounds were shown not to have any considerable activity against Stigmasterol has not shown antiproliferative activity against C6 and HeLa cells. Hovewer, cytotoxic effect against human monocyte THP1-Blue of this compound has defined in the literature (Hwang et al 2014). The anticancer activity of linarin which has several biological activities such as antianalgesic and anti-inflammatory (Jordon-Thaden and Louda 2003;Lim et al 2008) was determined to be inactive against C6 and HeLa cells.…”

“…TLR signaling pathways include activation of NF-κB and MAPK signaling. Its inhibitory effect on NF-κB and MAPK signaling could inhibit TLR signaling; however, only TLR4 inhibitory effect was reported [27]. Based on the experimental results, synthetic compounds, such as the ssRNA (single-stranded RNA) mimic poly(I), were used to induce TLR3 activity and NF-κB-mediated signal transduction pathway activity to identify a therapeutic agent for TLR-mediated diseases.…”
