cInstituto de Ecología, Universidad del Mar, 70902 Puerto Ángel Oax., México Soft Independent Modeling of Class Analogy (SIMCA) e Regressão por Mínimos Quadrados Parciais foram usados nesse trabalho para a identificação e quantificação de tiamina, riboflavina, nicotinamida e piridoxina, por espectrofotometria UV-Vis, sem realizar os procedimentos analíticos de separação e pré-concentração. Para a quantificação, os intervalos de trabalho estabelecidos foram 1-14 mg L -1 para riboflavina, 2-26 mg L -1 para tiamina, 2-30 mg L -1 para nicotinamida, e 2-22 mg L -1 para piridoxina. Os resultados de recuperação obtidos foram superiores a 95% em todos os casos, para a análise de amostras sintéticas e comerciais. Na busca de cada vitamina alvo, foi construído um modelo de classificação com duas categorias: (i) com a vitamina de interesse e (ii) sem a vitamina de interesse. A capacidade discriminatória de cada modelo de classificação foi avaliada para aprendizagem, para um conjunto de amostras teste e para amostras comerciais, com resultados satisfatórios, com exceção da riboflavina. Assim, um método simples e confiável é proposto para a estimativa simultânea desses compostos.Soft Independent Modeling of Class Analogy and Partial Least Squares Regression were used in this work for the identification and quantification of thiamine, riboflavin, nicotinamide and pyridoxine by UV-Vis spectrophotometry, without separation or preconcentration steps in the analytical procedure. For quantitative purposes, the working range established was 1-14 mg L -1 for riboflavin, 2-26 mg L -1 for thiamine, 2-30 mg L -1 for nicotinamide, and 2-22 mg L -1 for pyridoxine. Recovery results higher than 95% were obtained in all cases during the analysis of synthetic and commercial samples. In the screening of each target vitamin, a classification model was built with two classes: (i) with the vitamin of interest, and (ii) without it. The discriminate capability of each classification model was evaluated for learning, independent testing and commercial samples, resulting in satisfactory findings with exception of riboflavin. Thus, a simple and reliable method is proposed for the simultaneous estimation of these compounds.

Recent years have seen an increased interest in DNA trace detection methods involved in many areas of bioanalytical research, such as quantitation of genetically modified (GM) ingredients in food products. There is little in the way of standardisation of data handling from these methods, and the data generated needs to be analysed appropriately if the results are to be interpreted correctly. This paper describes particular aspects of real-time PCR trace detection methods in order to increase the understanding of data generated using this bioanalytical technique. Using the specific example of GM soya detection and quantitation, it focuses on the production of calibration curves based on the mean and individual data values, the interpretation of correlation coefficients, regression techniques, and discusses suitable data analysis arising from simple and more complex experimental designs following transformation. By using the approaches outlined in this paper, more accurate analysis of data from real-time PCR and GM trace detection methods could be achieved.

In this work, the artificial neural networks (ANN) and partial least squares (PLS) regression were applied to UV spectral data for quantitative determination of thiamin hydrochloride (VB1), riboflavin phosphate (VB2), pyridoxine hydrochloride (VB6) and nicotinamide (VPP) in pharmaceutical samples. For calibration purposes, commercial samples in 0.2 mol L -1 acetate buffer (pH 4.0) were employed as standards. The concentration ranges used in the calibration step were: 0.1 -7.5 mg L -1 for VB1, 0.1 -3.0 mg L -1 for VB2, 0.1 -3.0 mg L -1 for VB6 and 0.4 -30.0 mg L -1 for VPP. From the results it is possible to verify that both methods can be successfully applied for these determinations. The similar error values were obtained by using neural network or PLS methods. The proposed methodology is simple, rapid and can be easily used in quality control laboratories.Keywords: water-soluble vitamins; UV spectrophotometry; multivariate calibration. INTRODUÇÃOVitaminas são substâncias orgânicas indispensáveis à manutenção das funções metabólicas do organismo. A maior parte das vitaminas atua como coenzima; isto explica seu papel essencial nos processos biológicos. Essas substâncias apesar de essenciais ao metabolismo, não são sintetizadas pelo organismo e, em virtude disso, necessitam ser adquiridas de outras formas 1 . Em dias atuais, alimentos enriquecidos e medicamentos estão se tornando importantes meios para a obtenção de vitaminas para o nosso organismo. Dessa forma, métodos que assegurem a qualidade destes produtos são essenciais. A maioria dos métodos descritos na literatura envolve separações químicas e físicas 2-5 . Dentre esses métodos, incluem-se os de cromatografia líquida de alta eficiência (CLAE) e os que envolvem titulação. As principais desvantagens destes métodos consistem em um relativo alto custo, emprego de reagentes tóxicos e, em muitos casos, um grande consumo de tempo para a execução das análises. Desde a ultima década, contudo, o uso de métodos espectroscópicos aliados à calibração multivariada tem-se mostrado um meio rápido e eficiente para a determinação de diversos tipos de substâncias em amostras farmacêuticas. Algumas publicações mostram o uso de téc-nicas espectrofotométricas e eletroquímicas em conjunto com mé-todos de calibração multivariada para a determinação de vitaminas [6][7][8][9][10] .
