Single ether group in a glycol-based ultra-thin layer prevents surface fouling from undiluted serum

Abstract: Through systematic structural modification, it is shown that the internal, single oxygen atom of simple monoethylene glycol-based organic films is essential for radically altering the fouling behaviour of quartz against undiluted serum, as characterized by the electromagnetic piezoelectric acoustic sensor. The synergy is strongest with distal hydroxyls.

“…4. In comparison, plasma adsorption on bare quartz caused a much greater shift in resonant frequency of Δf $ À28 kHz, close to the value recorded for full serum (Δf ¼ À30.9 73.7 kHz) (Sheikh et al, 2012). Of higher importance for LPS biosensing is the entirely different specific response profile obtained with plasma samples spiked with high dose of LPS (1000 pg/mL), an endotoxemia level of the order of magnitude that can be found in patients in dire need of treatment (Opal et al, 1999) such as those admitted to intensive care units (Marshall et al, 2002).…”

“…SI-4). With respect to hemocompatibility, the presence of MEG-OH background areas -formed during PMB attachment (vide supra) -is certainly beneficial since this type of coating has been shown to display remarkable antithrombogenicity (Fedorov et al, 2014), in addition to pronounced antifouling properties (Sheikh et al, 2012) ( Fig. SI-3).…”

“…Sensing relies on the acoustic wave physics of our electromagnetic piezoelectric acoustic sensor (EMPAS), a highly sensitive flow-injection device able to perform measurements in a real-time and label-free manner at tunable frequency. [14,[18][19][20][21][22][23] Biosensing properties are evaluated and discussed in terms of avidin specific and non-specific adsorption (NSA). Sacrificial bovine serum albumin (BSA) is used to block NSA.…”
