Spatial modeling of prostate cancer metabolic gene expression reveals extensive heterogeneity and selective vulnerabilities

Abstract: Spatial heterogeneity is a fundamental feature of the tumor microenvironment (TME), and tackling spatial heterogeneity in neoplastic metabolic aberrations is critical for tumor treatment. Genomescale metabolic network models have been used successfully to simulate cancer metabolic networks. However, most models use bulk gene expression data of entire tumor biopsies, ignoring spatial heterogeneity in the TME. To account for spatial heterogeneity, we performed spatially-resolved metabolic network modeling of the… Show more

“…The increased fatty acid uptake in all aggressive tumor lines suggests a metabolic switch associated with the use of exogenous fatty acids in the spread of metastatic disease. This idea is supported by previous studies implicating fatty acid uptake and oxidation as a key promoter of tumor cell migration [70,75,76].…”

“…Along with increased fatty acid metabolism reportedly correlated with increased metastatic and aggressive tumors, tumor metabolic heterogeneity has also been reported in the literature to correlate with increased tumor aggressiveness [ 75 ]. Previous work highlights the presence of lipid spatial heterogeneity, specifically, and the importance of capturing this metric to better phenotype the tumor metabolic landscape [ 76 ]. Further, increased heterogeneity captured via texture analysis has previously been associated with increased tumor grade and metastases [ 77 , 78 ].…”

“…A number of other pathologies have also been associated with SLCO2A1. Thus, recent studies have provided new evidence for the roles of the SLCO2A1 protein in carcinogenesis [160,[162][163][164][165][166], in experimental colitis [167], in the parturition process [168], in cigarette-smoke-induced lung inflammation [169], in a gastroesophageal reflux disease [170], and in the non-healing diabetic foot ulcer [171]. The role of the ion channel function of SLCO2A1 in the mechanism of development of these pathologies remains to be elucidated.…”

“…Heterogeneity in the spatial distribution of metabolites within the tumor microenvironment (including endogenous, immunosuppressive, and drug metabolites) also plays a crucial role in influencing gene expression and determining the cellular response to therapies [ 27 ]. Metabolic profiling of native tissues in a spatially resolved manner is more challenging than spatial RNA profiling, as amplification of the metabolite signal is limited and labeling toolkits are lacking.…”
