Standardization of Functional Reporter and Antibiotic Resistance Cassettes to Facilitate the Genetic Engineering of Filamentous Fungi

Abstract: The unique physiological properties of fungi are useful for a myriad of applications, which could greatly benefit from increased control of native pathways and introduction of recombinant genes. However, fungal genetic engineering is still limited in scope and accessibility, largely due to lack of standardization. To help standardize the genetic engineering of filamentous fungi, we created BioBricks of commonly used antibiotic resistance genes, neomycin phosphotransferase (nptII) and hygromycin phosphotransfer… Show more

“…Bacteria are single celled, whereas most fungi are multicellular; bacteria have three distinct shapes, while fungi have various shapes which lead to mycelium formation; bacteria reproduce sexually, whereas fungi are capable of reproducing both sexually or asexually. All these differences make fungi more durable and resistant against some antibiotics [ 49 ]. Until now, research has tended to focus on antibacterial nanoparticles rather than on nanoparticles against fungi.…”

“…Finally, genes conferring resistance to antibiotics have been widely used as markers for the selection of transformed cells. The most widely used marker system is the antibiotic resistance to hygromycin [ 310 , 311 ], but also other antibiotics markers are used, such as bialaphos/phosphinothricin, geneticin/neomicyn, phleomycin and kanamycin resistance [ 312 , 313 , 314 ].…”

“…Bacteria are unicellular, are of three discrete shapes, and reproduce sexually, while most fungi are multicellular, exhibit a wide range of shapes that can lead to mycelium formation, and possess the ability to reproduce both sexually and asexually. These distinctions make fungi more vigorous and resistant to certain antibiotics . Research efforts have predominantly concentrated on antibacterial NPs, with limited attention given to NPs against fungal infections.…”
