Structural characterization and SARS-CoV-2 inhibitory activity of a sulfated polysaccharide from Caulerpa lentillifera

Abstract: Caulerpa lentillifera (Bryopsidophyceae, Chlorophyta) is an edible seaweed attracting great attention for its expansion of farming scale and increasing consumption in these years. In the present study, a sulfated polysaccharide (CLSP-2) was isolated and separated from C. lentillifera , and its chemical structure was elucidated by a series of chemical and spectroscopic methods. Among these methods, mild acid hydrolysis and photocatalytic degradation were applied to release mono… Show more

“…The corresponding C-2, C-3, C-4, C-5 and C-6 were assigned to δ 71.07, 72.97, 66.82, 69.16 and 60.66 ppm. These results were consistent with the previous report by You et al [ 24 ].…”

“…C. lentillifera (100 g) was washed with tap water to remove sediments, and neutral protease (0.3%, w/w) was added to hydrolyze proteins at 50 °C for 2 h. Then, the water-soluble dietary fiber (CLP) was extracted by our previously reported method. 3 After that, the precipitate was resuspended in distilled water (100 mL) and centrifuged at 3000 g for 10 min to remove water-soluble materials, and this step was repeated three times. Then, the precipitation was resuspended in ethanol (100 mL) and centrifuged at 3000 g for 10 min to remove alcohol-soluble impurities with three repetitions.…”

“…The analyses for monosaccharide composition, Fourier transform-infrared (FT-IR) spectroscopy, and X-ray diffraction (XRD) of the CL-IDF were carried out according to our previous method. 3 Briefly, for monosaccharide composition, the CL-IDF was hydrolyzed by 2 M trifluoroacetic acid and labeled with 1-phenyl-3-methyl-5-pyrazolone (PMP). After that, the PMP derivative was analyzed by high-performance liquid chromatography (HPLC) equipped with a C18 column (250 mm × 4.6 mm, 5 μm, Dikma, CN) and a photodiode array detector (Thermo Fisher Scientific, Switzerland).…”

“…In addition, seaweed polysaccharides can inhibit viral transcription and replication through direct interference with viral replication enzymes or inhibition on other intracellular targets [ 140 ]. Marine polysaccharides, especially sulfated polysaccharides from marine algae, appear to block viral infection by inhibition of one or more of these stages, with the specific mechanisms dependent on the structure of the individual polysaccharide [ 140 , 141 , 142 , 143 , 144 ].…”
