Studies with marked antisera. Quantitative studies with antisera marked with iodine 131isotope and their corresponding red-cell antigens

Abstract: MOLYBDATE AND HYDROLYSIS OF PHOSPHATES 745 3. It is suggested that the effect is due to three factors. (a) If magnesia mixture is added to the reduced phosphomolybdate complex and the resulting precipitate analysed colorimetrically, recoveries of inorganic phosphate are low. (b) Free molybdic acid is reduced, giving molybdenum blue. Contrary to prevailing opinions, molybdenum blue is extracted from acid solutions by i8obutanol. (c) The specific extinction coefficient is about 4-5 times higher when the colour o… Show more

“…The interpretation of the data obtained in these studies is based on the specificity of the I-131 bound to red cells (11)(12)(13). The effective exploitation of I-131 red cell isoantibodies depends on unequivocal evidence that the 1-131 taken up by the red cells represents the specific isoantibody and conforms with the known behavior of these antibodies.…”

“…Carrier-free 181I in weak thiosulphate solution was obtained from The Radiochemical Centre, Amersham, Bucks. Iodination was carried out by a modification of the method of Boursnell et al (1953) (Veall, Pearson & Hanley, 1955). The "8L1-labelled serum was dialysed against 500 vol.…”

“…Filitti-Wurmser, JacquotArmand, Aubel-Lesure & Wurmser (1954) computed thermodynamic constants for anti-B antibody against group B erythrocytes; antibody concentration was measured indirectly, in terms of the maximum number of erythrocytes that the antiserum could agglutinate. Boursnell, Coombs & Rizk (1953) used 1I as a protein label to investigate the uptake by red cells of the antibody anti-D.…”

“…Radioactive phosphorus was used in the form of a carrier-free phosphoric acid (pH 2 to 3) and mixed with the standard diluent used for the initial dilution. The method of Boursnell, Coombs & Rizk (1953) was used to iodinate the plasma proteins with radioactive iodine. The experi¬ ments with radioactive materials were carried out with the co-operation of TIME (HR) Text- fig.…”
