Study of acetylcholinesterase inhibitors using CE with contactless conductivity detection

Abstract: The hydrolysis of acetylcholine and acetylthiocholine as catalyzed by the enzyme acetylcholinesterase was monitored by CE with contactless conductivity detection by determining the acetate produced in the reaction. This approach eliminates the need for a color forming derivatization procedure. The effects of the three inhibitors galanthamine, hyperzine A and paraoxon on the enzyme kinetics could also be investigated by the new procedure and the IC(50) values were determined. The contactless conductivity detect… Show more

“…Importantly, synthetic inhibitors are used as medicines to treat neurodegenerative disorders such as Alzheimer's disease (AD). 19 Pharmacological treatment of AD is currently based on AChE inhibitors, such as tacrine, donepezil, rivastigmine, and galantamine, which maintain high levels of ACh at the muscarinic and nicotinic receptors in the central nervous system, exerting beneficial effects on cognitive, functional, and behavioral symptoms of the disease. 20 Therefore, AChE inhibition assay is utilized to screen for potent AChE inhibitors from edible resources such as black soybean seed coat.…”

“…However, the sensitivity of CE is a shortcoming because of the short light path. Hence, a variety of detection instruments are combined directly with CE, including laser‐induced fluorescence, ultraviolet absorbance, MS, and electrochemical detectors . In spite of all its assets, CE–Q‐TOF–MS remains rarely used in the determination of natural products.…”
