Synthesis and study of the complex formation of a cationic alkyl-chain bola amino alcohol with DNA: in vitro transfection efficiency

Abstract: Recent studies point out that bolaamphiphiles can be used in non-viral gene therapy. Due to their bipolar character, they may span a membrane and thus stabilize or destabilize it, which could be relevant for DNA transfer across a biological membrane. Since there are only very few studies on bolaamphiphile application in DNA transfection, it is difficult to assess whether they will bring additional advantages to the class of non-viral vectors. A bolaamphiphile with a hydrophobic chain of 22 carbon atoms with tr… Show more

“…In 2015, a structurally simple BA was introduced by Paiva et al ( 2015 ) that featured quaternary ammonium and primary alcohol head groups separated by a 22-carbon hydrocarbon linker. Here again, it was found that the BA can function as a standalone DNA binding material (bolaplex) or a component of a more complex formulation utilizing traditional cationic lipids such as 1,2-dioleoyl-3-trimethylammonium-propane (DOTAP).…”

“…18,194 The liposome of bolaamphiphiles (denoted as bolasome) has been explored as a nonviral vector because it has decent transfection efficiency and a large room to load genes, peptides, 195 or proteins, 196 while viral vectors can deliver only nucleic acids. 197,198 In addition, bolasomes offer diverse tailored activity such as specific cell recognition and enhanced transfection efficiency which are implanted through the peptide design. The robust bolasomes can be disassembled by external stimuli in a controlled manner, 187 which is particularly relevant for targeted delivery.…”

“…Bolaamphiphile is a promising constituent of gene carriers. Compared to the conventional liposomes made of phospholipids or other ionic lipids with a single hydrophilic head group, the bolaamphiphile assemblies display higher mechanical stability with the capability to fuse into the target cells. , The liposome of bolaamphiphiles (denoted as bolasome) has been explored as a nonviral vector because it has decent transfection efficiency and a large room to load genes, peptides, or proteins, while viral vectors can deliver only nucleic acids. , In addition, bolasomes offer diverse tailored activity such as specific cell recognition and enhanced transfection efficiency which are implanted through the peptide design.
