The repair of tissue defects after injury is of significant clinical and research importance. A variety of chitosan‐based hydrogels have been investigated and applied to address this problem. By using different synthetic methods, the hydrogels exhibit distinct physical properties, including porosity, adhesion, and stiffness. These properties are considered important factors affecting cell proliferation and tissue regeneration. Meanwhile, drug delivery and cell delivery are the two main strategies to improve the therapeutic effects of chitosan‐based hydrogels, but the choices of cells or drugs are quite varied and largely depend on the specificity of the treated tissues. The present review summarizes the physicochemical properties of chitosan‐based hydrogels and their influences on cells, and lists specific ways to promote the tissue regeneration in different systems of the human body. © 2018 Wiley Periodicals, Inc. In this paper, the micro-structured hydroxyapatite (HA)/TiO 2 coating was firstly produced on titanium surface by one-step micro-arc oxidation (MAO), and then bone morphogenic protein-2 (BMP-2)-encapsulated chitosan (CS) coating was prepared on the HA/TiO 2 surface by dip-coating method to endow the bioactive HA/TiO 2 coating with good antibacterial property and improved biological properties. The bonding strength between coatings was studied by scratch method. The degradability of CS, BMP-2 release behavior, bioactivity, biocompatibility and antibacterial property of the BMP-2/CS/ HA/TiO 2 composite coating were examined by in vitro tests. The results showed that, after loading BMP-2/CS on HA/TiO 2 surface, the surface roughness and wettability decreased but the bonding strength between the HA coating and titanium substrate was improved. CS, as a suitable carrier for sustained release of BMP-2, can control the release of BMP-2 continuously for 4 weeks long in a simulated human serum, and in PBS solution BMP-2 can be released more slowly. In addition, CS endowed the BMP-2/CS/HA/TiO 2 composite coating with good antibacterial activity. The BMP-2/CS/HA/TiO 2 showed good bioactivity, and it was also beneficial for cell adhesion, spreading and proliferation. Based on the above results, BMP-2/CS/HA/TiO 2 has great potential to be applied in clinic.

pH responsive chitosan and 3-Glycidyloxypropyl trimethoxysilane (GPTMS) hydrogels were synthesized by the sol-gel crosslinking reaction. GPTMS was introduced to influence several behaviors of the chitosan hydrogels, such as the swelling ratio, mechanical properties, swelling thermodynamics, kinetics, and expansion mechanism. The functional groups of Chitosan/GPTMS hybrid hydrogels were verified by FT-IR spectrometer. Differential scanning calorimetry (DSC) and the thermogravimetric analysis (TGA) were used to analyzed the thermal behavior of water molecules, the expansion of thermodynamics, and the content of water molecules in the hydrogel. The results show that hydrogel consists of 50 wt.% GPTMS (CG50) and has good mechanical properties and sensitivity to pH response characteristics in the acidic/alkaline buffer solution. The increase of GPTMS content leads to the increase of hydrophobic groups in the hydrogel and causes the decrease of the overall water content and the freezing bond water content. When the hydrogels were immersed in acid solution, the interaction force parameter was smaller than that of DI-water and alkaline. It means that the interaction forces between hydrogel and water molecules are relatively strong. The swelling kinetics of hybrid hydrogels were investigated to inspect the swelling mechanism. The result is consistent with the Fisk’s diffusion mechanism, meaning that the rate of water penetration is adjustable. The biodegradable hydrogel (CG50) in this study has good environmental sensitivity and mechanical properties. It is suitable to be applied in the fields of drug release or biomedical technology.
