Synthesis of oligosaccharide fragments of the Streptococcus pneumoniae type 14 capsular polysaccharide and their neoglycoconjugates with bovine serum albumin

Abstract: 2 Aminoethyl glycosides of tetra , hexa and octasaccharide fragments of the bacteria Streptococcus pneumoniae type 14 capsular polysaccharide were synthesized within a project directed to the development of pneumococcal conjugated vaccine based on synthetic carbo hydrate ligands. Squarate method was used to obtain neoglycoconjugates of the synthesized oligosaccharides with bovine serum albumin.

“…Oligosaccharides were conjugated to BSA (Sigma-Aldrich, USA) and biotin (structures and designations are given in Figure 1 ), as previously described ( 22 , 23 ). BSA has been frequently used as a protein carrier in engineered immunogenic glycoconjugates and other types of biomolecular systems ( 24 ).…”

“…BSA has been frequently used as a protein carrier in engineered immunogenic glycoconjugates and other types of biomolecular systems ( 24 ). Matrix laser desorption ionization time-of-flight data demonstrate that the tetra-BSA, hexa-BSA, and octa-BSA conjugates contained on average 11, 15, and 9 OS ligands per protein molecule, corresponding to carbohydrate contents (by weight) of 10, 19, and 16%, respectively ( 22 ).…”

“…The HS conjugate [β-Gal-(1 → 4)-β-Glc-(1 → 6)-[β-Gal-(1 → 4)]-β-GlcNAc-(1 → 3)-β-Gal-(1 → 4)-β-Glc-O-spacer] 15 -BSA was prepared by the squarate method and contained, according to MALDI-TOF data, on average, 15 HS residues as previously described ( 30 , 31 ) and 19% carbohydrates on a dry weight basis (Figure 1 ). The BSA is often used as a protein carrier to design immunogenic GCs and biomolecular systems ( 32 ) of other types.…”

“…In addition to discovering new possibilities in the synthesis of well‐defined glycoconjugates consisting of already known components, some studies, more biochemical than nanotechnological, have focused on immunogenic efficiency and the synthesis of new poly‐/oligo‐saccharides, mainly derivatives of bacterial cell wall epitopes . Similarly, Johannes et al.…”
