Tannic Acid-Mediated Surface Functionalization of Polymeric Nanoparticles

Abstract: Polymeric nanoparticles (NPs) are decorated with various types of molecules to control their functions and interactions with specific cells. We previously used polydopamine (pD) to prime-coat poly(lactic-co-glycolic acid) (PLGA) NPs and conjugated functional ligands onto the NPs via the pD coating. In this study, we report tannic acid (TA) as an alternative prime coating that is functionally comparable to pD but does not have drawbacks of pD such as optical properties and interference of ligand characterizatio… Show more

“…55 Briefly, 100 mg of PLGA and 3.6 or 0.5 mg of DiR were dissolved in 10 mL dichloromethane (DCM). The polymer solution was added to 30 mL of 5% PVA solution and emulsified with a Vibra-Cell probe sonicator (Sonics, Newtown, CT, USA) at 40% amplitude with a 4s on and 2s off duty cycle for 2 min.…”

“…The contact angle did not vary upon further increases in the concentration of TA, indicating a possible saturated coating. The quantitative analysis of TA absorption was realized by a bicinchoninic acid (BCA) assay (Figure E) . A plot of net TA absorption mass to TA concentration shows that the TA absorption content increased with TA concentration up to 20 w/w%, and this, combined with the results of contact angle and WOA measurements, suggests that a monolayer coating gives way to a multilayer one at higher TA concentrations.…”

“…TA Absorption Experiment : The absorption of TA on skin tissues was quantified using the BCA assay . A series of TA solutions were prepared at concentrations of 0%, 0.00032%, 0.0016%, 0.008%, 0.04%, 0.2%, 1%, 5%, 10%, and 20%, and concentration‐dependent absorption of TA was performed by spreading 100 µL of each solution on a square skin tissue (1 cm × 1 cm).…”

“…In contrast to PDA coatings, natural-inspired polyphenols coating is colorless, low-cost, and environment friendly [85]. A rapidly increasing report using polyphenol coatings for various purposes has been witnessed [86][87][88] (recently reviewed by Ejima et al [89]). However, naturalinspired polyphenol coating has not been applied in the field of CE so far, so natural-inspired polyphenol coatings used in CE may be a promising topic in the future.…”
