Targeting of the signal transducer Smo links microRNA‐326 to the oncogenic Hedgehog pathway in CD34+ CML stem/progenitor cells

Abstract: Aberrant expression and function of microRNAs (miRNAs) in leukemia have added a new layer of complexity to the understanding of development and progression of the disease state. However, their targeting of specific signaling pathways responsible for the maintenance and survival properties of leukemic stem cell (LSC) still remains to be further clarified. Hedgehog (Hh) signaling, a highly conserved developmental pathway, has been proven as a functional pathway for LSCs, and loss of this pathway impairs the deve… Show more

“…miR-152 is crucial for anti-tumor effect of nature killer-cells by upregulating HLA-G (Bian et al, 2015). Down-regulation of miR-326 promotes cell proliferation in CML CD34 + cells (Babashah et al, 2013). Silencing of miR-708 could promote NF-kB signaling in CLL (Baer et al, 2015).…”

“…For example, Suppressor of fused ( Su(fu) ) is targeted by miR-214 in Zebrafish to enable precise specification of muscle cell types by sharpening cellular responses to Hh (Flynt et al, 2007). In non-small cell lung cancer cell line, miR-212 acts as oncogene by targeting PTCH1, human homolog of Drosophila Ptc, to increase cell proliferation, migration and invasion (Li et al, 2012), while in chronic myeloid leukemia (CML), miR-326 targets Smo to decrease cell proliferation and increase apoptosis (Babashah et al, 2013). …”

“…This suggests a miR-326-mediated negative feedback loop in modulating Shh pathway during lung development (Figure 2) (106). In brain tumor progenitor cells, miR-326 was also shown to modulate Shh signaling by targeting smoothened (107,108).…”
