Taxonomic study of the genus Tepidiphilus: transfer of Petrobacter succinatimandens to the genus Tepidiphilus as Tepidiphilus succinatimandens comb. nov., emended description of the genus Tepidiphilus and description of Tepidiphilus thermophilus sp. nov., isolated from a terrestrial hot spring

Abstract: Comparative phenotypic, chemotaxonomic and genetic analysis revealed significant similarities among strains of the genera Tepidiphilus and Petrobacter . Analysis of 16S rRNA gene sequences and DNA–DNA relatedness of the type strains Tepidiphilus margaritifer … Show more

“…Although the type species was first reported in 2003, this genus only comprises three species at present: Tepidiphilus margaritifer , Tepidiphilus succinatimandens and Tepidiphilus thermophilus . The isolation of these micro-organisms frequently occur in geothermal conditions, such as thermophilic water treatment sludge [1], petroleum reservoir [2] and a terrestrial hot spring [3], which favour specific metabolic microbial types. This may explain to some degree why such few species in this genus has been discovered.…”

“…Preparation of fatty acid methyl esters, gas chromatographic separation, analysis and identification of peaks were performed by a method described earlier (Kumari et al 2014b). Polar lipids were extracted from 200 mg freeze dried cells following the method described by Bligh and Dyer (1959), separated by two dimensional TLC on silica gel 60 F 254 plate (Merck, Germany) and analyzed by spraying with appropriate solvents (Poddar et al 2014). Solvent systems used in this study were taken from Kumari et al (2014a).…”

“…Amplification, sequencing and secondary structure based analysis of 16S rRNA were performed as described by Poddar et al (2014) where the 16S rRNA of Esherichia coli (accession no J01695) was used as the reference secondary structure template. The primer and PCR conditions for amplification of 16S rRNA were adapted from de la Haba et al (2010a).…”
