TCF21 regulates miR-10a-5p/LIN28B signaling to block the proliferation and invasion of melanoma cells

Abstract: Background and aim Some research has suggested that miRNA-10a (miR-10a-5p) had an inhibitory function in proliferation and invasion of cancers. Whereas the role of miR-10a-5p in melanoma has not been fully explored. This study aims to confirm LIN28B as the targeted gene of miR-10a-5p which was explored in melanoma cells. In addition, upstream regulatory molecule of miR-10a-5p was also investigated in melanoma cells. Methods Real-time Quantitative polymerase chain reaction (RT-qPCR) was adopted to analyze miR… Show more

“…In vitro studies overexpressing miR-10a-5p showed potentially cancer type specific effects on cellular proliferation. For example, overexpression of miR-10a in melanoma, acute myeloid leukemia, and laryngeal squamous cell carcinoma cells decreased cellular proliferation in these cancers ( 108 110 ).…”

“…Thus, we delved into the molecular mechanism of miR-10a-5p in porcine fat metabolism, revealing the role of miR-10a-5p in preadipocyte differentiation by targeting the KLF11 gene. Previous research has shown miR-10a-5p's involvement in inhibiting chicken granulosa cell proliferation by targeting MAPRE1 to suppress CDK2, as well as suppressing melanoma cell proliferation and migration [32,33]. It also impacts keratinocyte proliferation and directly targets hyaluronan synthase 3 [34].…”

“…MiR-10a-5p is directly regulated by TCF21, which binds to its promoter region. The TCF21/miR-10a-5p/LIN28B pathway could be targeted by a repressor in a new melanoma treatment in order to stop melanoma progression [ 95 ].…”
