BACKGROUND: The proportion of antituberculosis-induced liver injury on tuberculosis patients showed a relatively high incidence in many countries. Hepatic damage induced by antituberculosis drugs might potentially fatal. Isoniazid (INH) and rifampicin (RIF) are two main components of antituberculosis with crucial role for the incidence of liver injury. One of the herbal remedies that pose hepatoprotective action is purple sweet potato. Hepatoprotective action of purple sweet potato extract has been proved to pose antioxidant and anti-inflammatory activity. AIM: This study was designed to analyze the effect of purple sweet potato extract on rat interleukin-6 (IL-6) and tumor necrosis factor-α _(TNF-α) level, as well as liver histopathology feature in hepatic injury induced by INH-RIF. METHODS: This study was a randomized posttest-only control group design. Male Wistar rats (Rattus norvegicus), age 8–12 weeks, weight 180–220 g were included in this research (divided into three groups). Purple sweet potato extract was produced by maceration technique. IL-6 and TNF-α _level was measured by enzyme-linked immunosorbent assay, whereas liver histopathology examination was performed with hematoxylin and eosin staining. Statistical analysis was performed using one-way ANOVA and post hoc test. RESULTS: Liver IL-6 level in the normal, control, and treatment groups was 2.272±0.473, 3.315 ± 0.536, and 2.548 ± 0.304, respectively (p < 0.001). Liver TNF-α _level in the normal, control, and treatment groups was 26.476 ± 1.681, 48.584 ± 1.359, and 32.547 ± 1.528, respectively (p < 0.05). Histopathology feature of the liver in the control group showed significant liver congestion, liver degeneration, liver necrosis, and infiltration of inflammatory cell. Otherwise, histopathology feature of the liver in the treatment group showed minimal lesion. CONCLUSIONS: Purple sweet potato ethanol extract lowered the liver concentration of IL-6 and TNF-α, as well as improving liver damage in Wistar rats induced with isoniazid and rifampicin.

Objective: Iimmunomodulatory effects of Dawa-Ul-Kurkum, a Unani polyherbal preparation and the possible mechanisms in experimental model of paracetamol induced liver damage in rats. Materials and methods: The drug Dawa-Ul- Kurkum has been prepared and provided by CRIUM, Hyderabad. Silymarin was purchased from Sigma-Aldrich (USA) and paracetamol (marketed by Cipla LTD) purchased from general pharmacy shop. Biochemical kits were purchased from ERBA Diagonostic Mannheim Gmbh. Serum alanine aminotransferase (ALT), serum aspartate aminotransferase (AST) and serum alkaline phosphatase (ALP) were estimated by Kinetic method of International Federation of Clinical Chemistry (IFCC), serum bilirubin and total protein were estimated by End Point assay as per the instruction of the Kit Manufacture’s manual.Immunoglobulin and cytokine levels were assessed by ELISA kit manual method. Delayed type hypersensitivity reaction by Institoris et al method. Lipid peroxidation is measured spectrophotometrically as 2-thiobarbituric acid-reactive substance (TBARS), Glutathione (GSH) levels were estimated by the method of Ellman and NOx concentrations were determined by using the Griess reaction as described previously by Tracey et al. Results: Liver damage was induced in Wistar rats by administration of paracetamol and the effects of various drug treatments were assessed on morphological, biochemical, immunoglobulin, cytokine level and histological markers of liver toxicity. In the vehicle treated experimental group, administration of paracetamol induced significant derangements in liver function as evidenced by increased levels of SGOT, SGPT, alkaline phosphatase bilirubin and decrease in total protein, and reductions in body weight and increased liver weights as compared to controls. Histopathological examination showed Periportal necrosis with haemorrhages in experimental control. Pretreatment with Dawa-Ul-Kurkum (DK, 250 and 500 mg/kg) and hydroalcoholic extract of DK (HA, 500 and 1000 mg/kg) showed protective effects against the paracetamol induced biochemical, immunoglobulin, cytokine, delayed type hypersensitivity reaction and histopathological derangements of liver function following paracetamol. Conclusion: Both DK and its 50% hydro-alcoholic extract were found to be effective against paracetamol induced liver damage as they significantly prevented the hepatotoxic damage induced in rats.

Background: Based on the reports, Aloe vera extract, a herbal medicine, has antimicrobial and antiinflammatory effects. Also, this medicinal plant has considerable effects on innate immunity through cytokine secretion which resist fungal infections such as Candida albicans. Moreover, Aloe vera is considered as immunomodulatory agent and regulate inflammatory responses. Materials and Methods: In this study, twenty Balb/c mice were divided into five groups: four groups of infected mice with Candida albicans treated with different doses of Aloe vera extract and one healthy control group. Then, macrophage vital activity was measured by NBT assay. Nitric Oxide (NO) production as representative of macrophage activity was assayed by MTT test. Tumor Necrosis Factor alpha (TNF-α) and Interleukin-12 (IL-12) cytokines were measured by ELISA assay. Results: Based on our study results, macrophage vital activity and NO production decreased significantly. However, TNF-α and IL-12 cytokines increased significantly in mice model infected by Candida albicans. Conclusion: Aloe vera extract could enhance macrophage activity through increase in TNF-α and IL-12 cytokines in Candida albicans infections. Therefore, Aloe vera could stimulate innate immune responses to eliminate Candida albicans.
