The effects of FK1706 on nerve regeneration and bladder function recovery following an end-to-side neurorrhaphy in rats

Abstract: BackgroundImmunophilin ligands are neuroregenerative agents binding to FK506 binding proteins, by which stimulate recovery of neurons in a variety of injury nerves. FK1706 is a novel immunophilin ligand which has neuroprotective and neuroregenerative effects but without immunosuppressive activity. At present, most reports about FK1706 in ameliorating nerve injury and functional recovery are limited to cavernous nerve injury and erectile function recovery. This study aimed to demonstrate the effects of FK1706 o… Show more

“…During spinal cord injury, mesenchymal stem cells can be injected into the spinal cord using the subarachnoid space. Studies have shown that erythropoietin can accelerate this migration and repair spinal cord injury (16,17). The function of mesenchymal stem cells and their effect on spinal cord injury are shown in Figure 1.…”

“…A potentially promising discovery, that requires further investigation, is the utilization of FK1706 to improve post-ETSN nerve regeneration. Its potential benefit in neural regeneration derives from its capacity to raise the activation level of the Raf protein in nerve growth factor (NGF)induced cells in signaling pathways [15,16].…”

“…To circumvent immunosuppression and adverse immunosuppressive toxicity during treatment, the tacrolimus analogues GPI-1046, GPI-1485, and FK1706 were developed [44][45][46]. Studies have shown that FK1706 potently activates the MAPK pathway of nerve growth factor (NGF) and has significant neurotrophic effects by binding FK506 binding protein-52 (FKBP-52) and phospholipase C (PLC) to block inhibition of the MAPK pathway [47].…”
