The Fine Structure of Protein Bodies in Lupine Cotyledons during the Course of Seed Germination

Immunolocalization of alkaloids in lupin seeds (Lupinus spp.) has been performed by cryofixation and conventional methods. Alkaloids were localized in the protein bodies of the cotyledon cells. Some immunogold particles in the walls of these cells were also observed. There were no differences in the sites of localization between the two mentioned methods. X-ray microanalysis of elements showed the presence of P, Mg, S, and K in the protein bodies of cotyledon cells in lupin seeds. The role of K+ in alkaloids transport is discussed.

Protein was extracted into water (pH 8.0,l h) from defatted lupin seed kernel particles (mean 8,1425 pm). Intact and extracted particles were examined by scanning and transmission electron microscopy. The inner surface of the cell wall initially showed several small holes considered as possible sites of plasmodesmata. During extraction, protein bodies first became disrupted into smaller microstructures and then protein dissolved. Wide passages were formed between cells which allowed transport of protein. The molecular weight of proteins in solution did not change with time, suggesting that bulk flow rather than selective transport predominated.
