The Function of MoGlk1 in Integration of Glucose and Ammonium Utilization in Magnaporthe oryzae

Abstract: Hexokinases are conserved proteins functioning in glucose sensing and signaling. The rice blast fungus Magnaporthe oryzae contains several hexokinases, including MoHxk1 (hexokinase) and MoGlk1 (glucokinase) encoded respectively by MoHXK1 and MoGLK1 genes. The heterologous expression of MoGlk1 and MoHxk1 in Saccharomyces cerevisiae confirmed their conserved functions. Disruption of MoHXK1 resulted in growth reduction in medium containing fructose as the sole carbon source, whereas disruption of MoGLK1 did not c… Show more

“…Conidia were harvested with double-distilled water and filtered through with one layer Miracloth (Calbiochem, San Diego, CA, USA). Conidial germination and appressorial formation were observed on Gel Bond film (FMC Bioproducts, Rockland, ME, USA) as described previously (Zhang et al, 2011c;Chen et al, 2014). At least 100 conidia per replicate were examined.…”

“…Mycelia were collected from 2-day-old liquid culture for the measurement of biomass, and then washed by distilled water and lyophilized. Fungal biomass was determined by mycelial dry weight (Zhang et al, 2011c). All the experiments were repeated three times, each with three replicates.…”

“…However, the Δmohex1 mutant was indistinguishable from the wild‐type in its ability to catabolize GlcNAc. This may be due to the functional redundancy as several hexokinases are known to be present in M. oryzae (Zhang et al ., ) (http://onlinelibrary.wiley.com/doi/10.1111/1462-2920.13215/suppinfo).…”
