The location of GABAB receptor binding sites in mammalian spinal cord

Abstract: GABAB binding sites in rat spinal cord have been detected by receptor autoradiography using 3H-GABA in the presence of isoguvacine. The sites could be demonstrated throughout the spinal cord grey matter. The maximum concentration of GABAB sites occurred in lamina II with substantial amounts in other laminae of the dorsal horn. Much lower levels were detected in the ventral horn. Unilateral rhizotomy reduced the number of GABAB sites in the dorsal horn without affecting levels in the ventral horn. The greatest … Show more

“…GABA B receptor immunoreactivities are distributed in the laminae I-III in the spinal cord and in DRG neurons (Price et al, 1984;Price et al, 1987;Towers et al, 2000), consistent with data showing high density of binding sites for the GABA B agonist baclofen in these areas (Gehlert et al, 1985). Also, the mRNA of all three GABA B subunits is expressed in both the spinal cord and DRG (Towers et al, 2000).…”

“…GABA B receptors are present in the primary afferent neurons, spinal cord, and brain (Price et al, 1987;Chu et al, 1990;Towers et al, 2000). “…1B). Electrophysiological (Dutar & Nicoll, 1988;Calabresi, Mercuri, DeMurtas & Beradi, 1990) and anatomical (Price, Kelly & Bowery, 1987) evidence indicates that GABAB receptors can be located pre-andÏor postsynaptically. Thus we attempted to determine whether pre-andÏor postsynaptic mechanisms were involved in the baclofen-induced reduction of the visual response in the SC.…”

“…This binding accounted for a high percentage of GABA B sites in the dorsal horn (approximately 40% to 50% [102]), but less for GABA A (approximately 20% to 30% [101]). Dorsal rhizotomy produced a reduction in GABA B -binding sites in the superficial laminae comparable with that produced by neonatal capsaicin treatment, suggesting that these presynaptic GABA B receptors are preferentially localized to capsaicin-sensitive, small diameter myelinated and unmyelinated, typically nociceptive, afferents (103). Even though the GABA B receptor has recently been cloned (104), the precise location of the receptor in DRG neurons has yet to be investigated fully by in situ hybridization or immunohistochemistry.…”
