The power of AAA-ATPases on the road of pre-60S ribosome maturation — Molecular machines that strip pre-ribosomal particles

Abstract: The biogenesis of ribosomes is a fundamental cellular process, which provides the molecular machines that synthesize all cellular proteins. The assembly of eukaryotic ribosomes is a highly complex multi-step process that requires more than 200 ribosome biogenesis factors, which mediate a broad spectrum of maturation reactions. The participation of many energy-consuming enzymes (e.g. AAA-type ATPases, RNA helicases, and GTPases) in this process indicates that the expenditure of energy is required to drive ribos… Show more

“…by the Las1 endonuclease (Tang et al 2008;Kressler et al 2012;Schillewaert et al 2012;Talkish et al 2012;Gasse et al 2015). This study shows that Nog2, Nop53, and Rsa4 are present in pre-ribosomes when the N-terminal extension of L8 is deleted; however, we could not determine whether Las1 binding is affected.…”

“…This phase is also marked by the association of the Rix1 subcomplex and the AAA-ATPase Rea1 [21,33,99] (Figures 2N and 5). Interestingly, the interaction between Rix1 and Rea1 may be regulated, as indicated by a recent study with the orthologous human proteins showing that their association is controlled by a dynamic SUMO conjugation/deconjugation cycle [100].…”

“…First, the AAA-ATPase Drg1 releases the GTPase Nog1 and its binding partner Rlp24, thereby enabling recruitment of the r-protein eL24 [99,106]. Subsequently, Arx1 is dissociated by Jjj1 and Rei1 [103].…”

“…Many of these release events require GTPases or ATPases that are thought to drive conformational changes (Strunk and Karbstein 2009;Kressler et al 2012). The genetic analysis of these events in the later steps of 60S biogenesis reveals a special relationship between trans-acting factors and their devoted releasing factors (Lo et al 2010;Panse and Johnson 2010).…”
