The Protective Effects of Alisol A 24-Acetate from Alisma canaliculatum on Ovariectomy Induced Bone Loss in Vivo

Abstract: Alisma canaliculatum is a herb commonly used in traditional Korean medicine, and has been shown in scientific studies to have antitumor, diuretic hepatoprotective, and antibacterial effects. Recently, the anti-osteoclastogenesis of alisol A 24-acetate from Alisma canaliculatum was investigated in vitro. However, the influence of alisol A 24-acetate on osteoporosis in animals has not been investigated. The present study was undertaken to investigate the anti-osteoporotic effect of alisol A 24-acetate on bone ma… Show more

“…C57BL/6J mice (8 weeks) were purchased from Shanghai SLAC Laboratory Animal Co., Ltd, and housed five per cage under standard conditions (12 h light/12 h dark cycle, 21°C controlled temperature). For ovariectomized mice model, the mice were sham operated (sham group, n=9) or ovariectomized (OVX group, n=9) as previously described [ 48 ], four weeks after surgery, the mice were sacrificed and the bones were analyzed. For IL-1β-induced arthritis mice model, mice were anesthetized with 250 mg/kg intraperitoneal tribromoethanol (Sigma-Aldrich, St. Louis, MO) and knees shaved with scissors, and the patellar ligament visualized through a small skin incision.…”

“…TRACP is derived from osteoclasts and is a specific index reflecting bone resorption. ALP/TRACP can be used in evaluating bone turnover rate or bone remodeling [29]. We observed that bALP, TRACP and bALP/TRACP in serum and ALP, TRACP and ALP/TRACP in bone were significantly decreased in 17 m aged rats compared with 6 m rats ( Fig.…”

“…In general, bone specific alkaline phosphatase (BALP) is a marker of osteoblasts [ 55 ], and tartrate resistant acid phosphatase (TRAP) is a well-known marker of osteoclasts [ 56 ]. In a previous study, we showed that an estrogen deficiency induced by ovariectomy decreased the BALP and increased the TRAP activity [ 57 ]. Additionally, the administration of TSG to OVX mice increased the BALP levels and decreased TRAP.…”
