The role of the intestinal microbiota in the pathogenesis of host depression and mechanism of TPs relieving depression

Abstract: Depression is a prevalent neuropsychiatric disease with a high recurrence rate, affecting over 350 million people worldwide. Intestinal flora disorders and gut-brain-axis (GBA) dysfunction may cause mental disorders. Alterations in...

“…Dietary polyphenols have been found to act as neuroprotectants that possess pharmacological effects on the CNS for relieving anxiety and mitigating depression [125]. Robust evidence corroborates a bidirectional link between sleep disturbance and depression, also circadian dysregulation is one of the critical pathogenetic of depression nowadays.…”

“…The main mechanisms of polyphenols reducing depression are related to lowering oxidative stress and neuroinflammation, and increasing neurotrophin release. The processes proposed seem to also have an impact on serotonin, norepinephrine, and/or dopaminergic systems regulation to influence the HPA axis and modify the concentration of the BDNF [ 107 ], mitogen-activated protein kinase phosphatase-1 (MKP-1), and cAMP-responsive element-binding protein (CREB), and to normalize phosphorylation levels in the hippocampus, prefrontal cortex-modulating synaptic plasticity, activation of neuroplasticity, neurogenesis, and cell survival signaling pathways (PKA, CaMKII, PKC, MAPK/ERK, and PI3K) [ 108 ]. The use of dietary polyphenols to target various cell signaling pathways related to oxidative stress and inflammation, including MAPK, NFκB, and PI3K/Akt, is becoming a new method for preventing and treating depressive disorders.…”

“…The prevalence of depression in the general population during the COVID-19 outbreak is 25% ( 84 ). Alterations in the composition of the gut microbiota can increase the permeability of the gut barrier, activate systemic inflammatory and immune responses, modulate the release and efficacy of monoamine neurotransmitters, alter the activity of the hypothalamic-pituitary-adrenal axis and function, and alters the abundance of brain-derived neurotrophic factor (BDNF) ( 85 ). A deficiency of BDNF may lead to neuroplasticity impairment and depression.…”
