The role of the retinoblastoma protein (Rb) in the nuclear localization of BAG-1: implications for colorectal tumour cell survival

Abstract: Although the retinoblastoma susceptibility gene RB1 is inactivated in a wide variety of human cancers, the retinoblastoma protein (Rb) has been shown to be overexpressed in colon cancers, which is linked to the anti-apoptotic function of the protein. However, the mechanisms by which Rb regulates apoptosis are yet to be fully elucidated. We have established that Rb interacts with the anti-apoptotic BAG-1 (Bcl-2 associated athanogene-1) protein, and that a decrease in nuclear localization of BAG-1 is detectable … Show more

“…Conversely, ASAH-1, GPX2 and BAG-1 were down-regulated by EGC. BAG-1 encodes a known anti-apoptotic protein implicated in Bcl-2 signalling pathway [72,73]. ASAH-1 encodes acid ceramidase, an enzyme that catabolizes ceramide into sphingosine by deacylation.…”

“…The tumor cells with low proliferation might be more resistant to radiation and chemotherapy. Furthermore, the roles of overexpressed RB1 in suppressing the apoptosis might result in resistance to therapeutic radiation or chemotherapy, too [35, 36]. …”

“…The Saos-2 cells and MG-63 cells showed different results. Anti-apoptotic functions of Rb contribute significantly to the genesis and progression of tumors (Dasgupta et al, 2006) though the mechanisms by which Rb regulates apoptosis are yet to be fully elucidated (Clemo et al, 2005). The anoikis of suspended Saos-2 increased in a time-dependent manner reaching 72.2% cell death at 72 h of culturing.…”
