This study aimed at performing and comparing third molar development staging in extracted teeth (EX), in panoramic radiographs (PAN) and in cone beam computed tomography (CBCT). Extracted third molars (n=158, 95 maxillary, 63 mandibular) from 102 patients (36 males, 66 females) having at least one preoperative PAN and one CBCT volume were studied. Third molar development staging was performed in PAN, EX and in CBCT using Gleiser et al. (1955) technique modified by Köhler et al (1994). A polytomous logistic regression model was used to compare the staging performed EX and in CBCT with the gold standard staging in PAN. In all the pair-wise comparisons between staging modalities 100 third molars (63.3%) were equally staged. The other 58 third molars (36.7%) were staged with a maximum difference of one stage. No statistically significant differences between the three staging modalities were detected (p=0.26). The comparison between EX and PAN staging revealed higher similarity (p=0.98 in stages 5-10) than the comparison between CBCT and PAN staging (p=0.81 in stages 5, 7 and 9, and 0.80 in stages 6, 8 and 10). The studied third molar staging technique originally designed in PAN can be applied for third molar staging EX and in CBCT.

Objetivo: Realizar um levantamento dos artigos científicos que utilizaram acervos de imagem como fonte de pesquisa em Odontologia Legal e discutir essa utilização sob perspectivas éticas, bioéticas e legais. Material e Métodos: Foi realizada uma busca de artigos científicos nas bases de dados Lilacs, PubMed e Scielo entre os anos de 2015 e 2019 utilizando descritores nos idiomas português, inglês e espanhol. Foram incluídos apenas artigos da área de Odontologia Legal que utilizaram acervos de imagem como fonte de informação. Foram excluídos estudos laboratoriais ou clínicos, arqueológicos ou antropológicos, relatos de caso ou artigos de revisão, estudos prospectivos ou que utilizaram exames de imagem não pertencentes a acervos. Resultados: Foram selecionados 171 artigos científicos. Desses, 22,80% (n=39) foram realizados por autores vinculados a instituições brasileiras, e 77,19% (n=132) mencionaram aprovação por comitê de ética em pesquisa ou instituição correspondente com (ou sem) necessidade de Termo de Consentimento Livre e Esclarecido (TCLE). Conclusão: Os acervos de imagem compreendem uma importante fonte de informação para pesquisas em Odontologia Legal dada a quantidade de publicações realizadas no período avaliado. A maioria dos artigos mencionou aprovação ética e respeito à privacidade dos participantes, conforme os requisitos estabelecidos para dispensa do TCLE.

Stem cells from the apical papilla (SCAPs) are promising seed cells for tissue regeneration medicine and possess the osteogenic differentiation potential. Wnt5a, a typical ligand of the noncanonical Wnt pathway, exhibits diverse roles in the regulation of osteogenesis. The transcriptional co-activator with PDZ-binding motif (TAZ, WWTR1) is a core regulator in the Hippo pathway and regulates stem behavior including osteogenic differentiation. This study aims to examine how Wnt5a regulates SCAPs osteogenesis and explore the precise mechanistic relationship between Wnt5a and TAZ. Methods: SCAPs were isolated from developing apical papilla tissue of extracted human immature third molars in vitro. ALP staining, ALP activity and Alizarin red staining were used to evaluate osteogenic capacity. Osteogenic-related factors were assessed by qRT-PCR or Western blotting. Additionally, the receptor tyrosine kinase-like orphan receptor 2 (ROR2) was detected by immunocytofluorescence staining and silenced by small interfering RNA to verify the function of Wnt5a/ROR2 in TAZ-mediated osteogenesis. And we constructed TAZ-overexpression and β-catenin-overexpression SCAPs generated by lentivirus to explore the precise mechanistic relationship between Wnt5a and TAZ. Results: Wnt5a (100ng/mL) significantly suppressed ALP activity, mineralization nodules formation, expression of osteogenic-related factors. Meanwhile, it decreased the expression of TAZ mRNA and protein. TAZ overexpression promoted osteogenesis of SCAPs while Wnt5a could block TAZ-mediated osteogenesis. Furthermore, ROR2 siRNA (siROR2) was found to upregulate TAZ and canonical Wnt pathway signaling related molecules such as β-catenin, GSK3β and p-GSK3β. The suppression of Wnt5a/ROR2 on osteogenesis was significantly reversed by β-catenin overexpression through Wnt5a/ROR2/β-catenin/TAZ pathway. Conclusion:Taken together, the present study demonstrates that Wnt5a suppresses TAZ-mediated osteogenesis of SCAPs and there may be a Wnt5a/ROR2/β-catenin/TAZ pathway regulating osteogenesis of SCAPs. Moreover, Wnt5a could be a candidate for regulators in tissue regeneration.
