Thrombospondin-1 and CD47 regulation of cardiac, pulmonary and vascular responses in health and disease

Abstract: Cardiovascular homeostasis and health is maintained through the balanced interactions of cardiac generated blood flow and cross-talk between the cellular components that comprise blood vessels. Central to this cross-talk is endothelial generated nitric oxide (NO) that stimulates relaxation of the contractile vascular smooth muscle (VSMC) layer of blood vessels. In cardiovascular disease this balanced interaction is disrupted and NO signaling lost. Work over the last several years indicates regulation of NO is … Show more

“…5, one can also notice thrombospondin 1 (THBS1), sphingosine-1-phosphate receptor 1 (S1PR1, endothelial differentiation gene 1), and collagens I and V, which seem to be significantly involved in the pathogenesis of early graft restenosis. THBS1 and S1PR1 are VEGFs-dependent proteins [29, 31]. They affect ECs in an opposite manner.…”

“…Even from the time of conception, TSP-1 may affect inhibition of vascular vasodilation and apoptosis of glomerular membrane. 6,7,9,15 A limitation of this study was that the blood pressure measurements were performed on only one occasion. A 24-hour ambulatory measurement of blood pressure would be more accurate.…”

“…Increased TSP-1 can inhibit NO, that in turn causes vasoconstriction, significantly increased glomerulosclerosis, glomerular matrix accumulation, podocyte injury, renal infiltration with inflammatory cells, and altered renal function parameters. [10][11][12][13][14][15] Nevertheless, a comparison between human and animal models of TSP-1 levels and blood pressure should be viewed cautiously since TSP-1 levels and blood pressure in animal models may differ from human models.…”
