Thymidine kinase in rat liver during development

Abstract: 1. The activity of thymidine kinase in rat liver supernatant decreased with development to a value in the adult that was 1% of that in the 17-day foetus. 2. The foetal enzyme was more stable than the adult to gel filtration on Sephadex G-25 at 0 degrees . 3. The greater stability of the foetal enzyme to incubation at 45 degrees was attributable to the presence of higher concentrations of nucleotides in foetal liver supernatant. 4. The K(m) values for foetal and adult enzymes were approx. 2.5mum- and 2.1mum-thy… Show more

“…The pH optimum of hepatic thymidine kinase from neonatal rats was determined to be 8.0 (not shown). This latter value agrees with that found for thymidine kinases from neonatal liver, spleen, thymus and many transplanted Morris and Novikoff hepatomas (Klemperer & Haynes, 1968;Sneider etal., 1969;Tayloretal., 1972;Bresnick &Thompson, 1965). The different pH optimum of cardiac-muscle thymidine kinase distinguishes it from thymidine kinases characterized from other tissues.…”

“…2). This value agrees well with that of Klemperer & Haynes (1968), who found the Km value of hepatic thymidine kinase to be 2.1 pM, and with Bresnick & Thompson (1965) The pH optimum for cardiac thymidine kinase activity was 9.0 in foetal, neonatal and adult hearts (Fig. 3).…”

“…Protein was determined by the method of Lowry (20) using bovine serum albumin as standard. Thymidine kinase was assayed by the method of Klemperer and Haynes (21) (26) was added and the radioactivity was determined in a scintillation counter.…”

“…The latter diet had been shown to produce significantly elevated levels of colonic mucosal TK activity in previous studies (Calvert & Reicks, 1988). TK activity was assayed using the method of Klemperer & Haynes (1968), similar in principle to that employed by McKenna et al (1988).…”
