Protective effect of acetylcysteine against thiamethoxam-induced hepatic and renal toxicity in rats

The biochemical, oxidative DNA damage, and the histological alterations associated with thiamethoxam (MX) exposure, a second-generation neonicotinoid broadly used in Egyptian agriculture, were assessed. Also, the role of N-acetylcysteine (NAC), (150 mg/kg/ day) on the adverse effect of MX was investigated. Rats were orally preserved with a sub-lethal dose (1/50 LD 50 ) of MX at 31.26 mg/kg/day, five doses/week for 28 days. The MX exposure resulted in a significant decrease in rats' body weight, protein concentration of both serum and urine, sera catalase (CAT), and glutathione peroxidase (GPx) activities compared to control. In addition increase in sera creatinine, urea, bilirubin, alkaline phosphatase (ALP), superoxide dismutase (SOD), and malondialdehyde (MDA) levels were observed. While the assessment of DNA damage revealed significant altitude in 8-hydroxy-2'-deoxyguanosine (8-OH-2DG) levels in both serum and urine samples. The present findings were supported by microscopic observation of liver and kidney tissues. Evidently, thiamethoxam can damage liver and kidney functions impaired the DNA, and cause histoarchitecture lesions in rats at the tested sublethal dose. In addition, NAC supplementation significantly attenuated of MX adverse effect which reflects its protective influence against MXinduced hepatic-nephrotoxicity.

Hyperplasia of Leydig cells in both 100 mg/kg SFX-exposed group and 500 mg/kg SFX-exposed group explain the increase in the testosterone hormone in these groups.…”

The study objective was to evaluate the potential reproductive toxicity of sulfoxaflor (SFX) insecticide in male Sprague Dawley rats. To attain these objectives, forty male Sprague Dawley rats of 10–12 weeks old were randomly divided into four equal groups; the 1st group was used as a control group; the other three groups were exposed to 25, 100, and 500 mg/kg body weight SFX by oral gavage for 4 weeks. Relative testicular weight, testosterone, FSH, LH, MDA, and GPx levels, sperm viability, sperm morphology, sperm DNA damage, and histopathological changes in testes, epididymis, and seminal vesical of these rats were investigated after 4 weeks. The results showed that SFX exposure resulted in a significant increase in FSH, LH, MDA, and GPx levels as well as the percentage of dead and abnormal sperms and DNA damage in rat sperms. Histopathological examination of testes established testicular degeneration with coagulative necrosis as well as the proliferation of interstitial connective tissue infiltrated with inflammatory cells with congestion of intertubular blood vessels in epididymis and degeneration of lining epithelium of seminal vesicles.

Many pesticides are used together or in a method, eventually resulting in multiple exposures. These mixtures can lead to unpredicted adverse health consequences in the exposed population. Numerous studies on individual risk assessments are available, but combined usage's toxicity is still to estimate. So, the current study investigated hepatotoxicity induced by exposure to deltamethrin (DLT) and dimethoate (DM) and their mixture in adult male albino rats. Methods: Forty adult male albino rats were randomized into five equal groups (n=8); Group I: control, Group II: the vehicle group received 1ml corn oil. Group III: received DLT (5 mg/kg b.w.), Group IV: received DM (20 mg/kg b.w.), Group V: received DLT (5 mg/kg b.w) and DM (20 mg/kg b.w). Treatments were orally gavaged once-daily dose for twelve weeks. Results: Separate DLT and DM exposure significantly induced an increase in serum liver enzymes, including aspartate aminotransferase, alanine aminotransferase, alkaline transferase, and lactate dehydrogenase, bilirubin, and liver malondialdehyde levels accompanied by a decrease in serum total protein, plasma cholinesterase enzyme, liver superoxide dismutase, catalase, and reduced glutathione levels. These biochemical alterations were supported by the lesions observed in histological sections, decreasing the expression of Bcl-2 and DNA damage and genotoxicity detected by the comet assay. Conclusion: Co-administration of DLT and DM aggravated hepatic dysfunction, exhausting the endogenous antioxidant status and down-regulating the expression of Bcl-2. Thus, these mixtures can increase the harmful effects of each compound on the liver.
