Timing mechanisms in neuronal pathfinding, synaptic reorganization, and neuronal regeneration

Abstract: Precise temporal control of neuro-differentiation and postdifferentiation events is necessary for the creation of appropriate wiring diagram in the brain. To make advances in the treatment of neurodevelopmental and neurodegenerative disorders, and traumatic brain injury, it is important to understand these mechanisms. Caenorhabditis elegans has emerged as a revolutionary tool for the study of neural circuits due to its genetic homology to vertebrates and ease of genetic manipulation. microRNA (miRNA), a ubiqui… Show more

“…In a rat model of a middle cerebral artery occlusion and subsequent reperfusion injury, let-7 family members were upregulated at the 48-hour reperfusion time point [ 72 , 74 ]. In C. elegancs, several neurons have shown increased levels of let-7 as they age, whereby let-7 upregulation contributed to the decline of aging neurons’ regeneration potential [ 75 ]. Let-7 microRNAs was downregulated in radiation-exposed neural granule cell progenitors, as well as in medulloblastoma [ 76 ].…”

“…Many extrinsic and intrinsic factors that act to promote or block neuronal regeneration after nerve injury have been identified [11,16,44,[52][53][54][55][56] , but little is known for the molecular connection between neuronal debris removal and neuronal regeneration. It was recently shown in C. elegans that after peripheral nerve injury, proximal axon debris is removed and axon regenerates.…”
