Topical Delivery of Ebselen Encapsulated in Biopolymeric Nanocapsules: Drug Repurposing Enhanced Antifungal Activity

Abstract: Aim: Ebselen (Eb) is an example of a repurposed drug with poor aqueous solubility which requires sophisticated delivery system such as nanoencapsulation in nanocapsules for topical application. Materials & methods: Eb-nanocapsules were examined for morphology, activity against Candida spp., cytotoxicity and skin permeation. Results: Eb-nanocapsules were active against skin-infecting Candida tropicalis, Candida albicans and Candida parapsilosis yeasts (minimal inhibitory concentration values were about 4-, 2-an… Show more

“…Drug loaded nano-capsules are especially useful for skincare and dermatological treatments because of their enhanced bioavailability in dermal cells. Ebselen (Eb) is an example of a repurposed drug with poor aqueous solubility which requires a sophisticated delivery system such as nano-encapsulation for topical application as a promising, safe and complementary alternative to the treatment of cutaneous candidiasis (Jaromin et al, 2018). Examples of commercially available cosmeceutical products are "Hydra flash bronzer" a facial skin moisturizer, "Soleil soft-touch anti-wrinkle sunscreen, " "Soleil instant cooling sun" and "primordiale optimum lip" produced by Lancôme R .…”

“…Caco-2 cells were grown in Dulbecco's modified Eagle's medium (DMEM) medium containing 20% FBS, 1% non-essential amino acids, GlutaMAX™ and antibiotics. Cell viability was determined as previously described [ 52 ] using the MTT assay [ 53 ]. Different doses of 2 , 3 , 6 , 12 dissolved in DMSO, CQ (in water) as well as unloaded and 3 -loaded (2.0 mg/ml) coarse o/w emulsions were added to each well, mixed and incubated for 24 or 48 h. Results were expressed as IC 50 values, or as the percentage of survival cells with respect to the control (not treated cells).…”

“…Experiments were performed as described by Lewińska et al [ 1 ] and Jaromin et al [ 29 ]. Full-thickness pig skins were cleaned with tap water, separated from the underlying cartilage, then the subcutaneous fat was cut and stored at −20 °C for up to 1 month after preparation to ensure skin integrity.…”
