Topical Retinaldehyde with Glycolic Acid: Study of Tolerance and Acceptability in Association with Anti-Acne Treatments in 1,709 Patients

Abstract: Background: Retinaldehyde (RAL), a key metabolite between vitamin A and retinoic acid, acts by modulating differentiation and proliferation of keratinocytes, which is of interest in acne lesions, mainly retentional lesions. Glycolic acid increases the exfoliation of corneocytes explaining its mild activity on retentional lesions. Thus, RAL and glycolic acid combined in the same product (Diacnéal®) have complementary activities which can be of interest for acne patients. The aim of this study was to … Show more

“…Given recent evidence that showed P. acnes‐ induced inflammatory response in patients with acne, which in turn was regulated by vitamin A (retinol is a derivative of vitamin A) and vitamin D, retinoids will continue to be important in acne management . Retinaldehyde, the direct precursor of retinoic acid, has been shown to be efficacious and safe when combined with erythromycin, and it has been shown to be effective and less irritating than other retinoids in a large study of more than 1000 patients …”

“…have shown the very good tolerance of RAL/GA in a large population of patients ( n = 1709). Even in association with another topical treatment, tolerance was very good; no cutaneous side‐effect was noted, leading to a very high compliance 13 …”

“…Its combination with zinc gluconate could be assessed since the latter salt has the capacity to reach the dermis and exert anti-inflammatory and antibacterial effects against Propionibacterium acnes when given by the oral route (53). The implementation of a clinical trial is feasible since the combination of oral zinc and copper gluconate and disulfiram is U.S. Food and Drug Administration approved to treat alcoholism.…”
