and used in both pink mutation assay and comet assay using cellular nuclei from Tradescantia staminal hairs. The observation of staminal hair was realized along eight days (6-14) after treatment), flowers produced day 14 after treatment were utilized done according to Underbrink. In previous reports on plants, were comet assay was used, breaking cellular wall and separating by centrifugation gradient are necessary. Here, nuclei from staminal hairs were obtained by squashing the cells (is not necessary to utilize to break special procedure cellular wall), collected using a nylon mesh of 80Mm and next the comet assay was applied. Student's T test was the statistical test used for analyzing the comet assay data. Results. Both assays showed a great sensitivity to the studied mutagens. A relationship between the dose-pink event and the dose-tail length was evident. Even ResumenObjetivo. Evaluar la genotoxicidad de N-nitroso dietilamina (NDEA), hidrazida málica (MH) y etil metano sulfonato (EMS), en núcleos de Tradescantia (clona 4430) por medio de la prueba del cometa y de la prueba de mutación rosa, en los pelos estaminales de la misma planta. Material y métodos. Las plantas de Tradescantia (clon 4430) fueron obtenidas del Laboratorio de Citogenética y Mutagénesis del Centro de ciencias de la Atmósfera de la Universidad Nacional Autónoma de México, tratadas con NDEA a 1, 5, 10 mM, MH a 1, 5, 10 mM y EMS a 15, 30 y 45 mM, y utilizadas en la prueba de mutación rosa y en la del cometa, en núcleos celulares de los pelos estaminales. En la primera, la lectura de los pelos estaminales se realizó de acuerdo con el método de Underbrink. En otros estudios, que han aplicado la prueba del cometa en plantas, existe la necesidad de romper la pared celular y separar los núcleos por gradiente de centrifugación; en este caso, los núcleos de las células de los pelos estaminales fueron extraídos por aplastamiento sin aplicar un procedimiento especial para romper la pared, colectados por filtración en una malla de nylon y sometidos a la prueba del cometa. La prueba t de Student se usó para analizar los datos obtenidos. Resultados. Ambas pruebas presentaron una gran sensibilidad a los mutá-genos estudiados y hubo una relación evidente dosis-eventos rosa / longitud de la cauda. ABSTRACT. The genus Lupinus is widely distributed. Its seeds are used for animal and human food, and Lupinus possesses pharmacological potential because of its high content of quinolizidine alkaloids and flavonoids; however, there is little available information about its genotoxicity. We used the comet assay and staminal nuclei of Tradescantia (clone 4430) to evaluate the in vitro genotoxicity of 4 concentrations (0.01, 0.1, 0.5, and 1.0 mM) of alkaloid extracts of Lupinus mexicanus and Lupinus montanus, flavonoids of L. mexicanus, and commercial sparteine; nitrosodiethylamine was used as a positive control and untreated nuclei were used as a negative control. All concentrations of L. mexicanus and L. montanus showed significant genotoxic activity (P ≤ 0.05). A similar behavior was observed for flavonoid extracts of L. montanus except the 1.0 mM concentration. Sparteine showed genotoxic activity only at 0.5 mM. The order of Genotoxicities of Lupinus extracts genotoxicity of the compounds studied was as follows: L. mexicanus > L. montanus > flavonoids of L. montanus > sparteine. There is evident genotoxic activity in the compounds that were studied, particularly at lower concentrations (0.01 and 0.1 mM). Given the limited information about the genotoxicity of the compounds of L. mexicanus and L. montanus, further studies are necessary.

RESUMOO crescimento populacional e o conforto gerado pelo progresso têm contribuído amplamente para o aumento da poluição do ar, fazendo com que o ar de vários centros urbanos apresente cada vez mais substâncias nocivas aos seres vivos. O presente estudo objetivou caracterizar a qualidade do ar no município de Taubaté, Vale do Paraíba-SP utilizando biomonitoramento com Tradescantia pallida (Rose) D. R. Hunt purpurea Boom (Commelinaceae). O estudo foi desenvolvido num período de 10 meses (setembro/2010 a junho/2011) em cinco localidades (Rodovia Presidente Dutra -tráfego veicular intenso; Estiva -área residencial; Parque Aeroporto -área industrial; Campus Bom Conselho -centro da cidade com alto fluxo veicular e Agronomia -área rural. Foram realizadas semanalmente coletas das inflorescências jovens das plantas expostas. Após a coleta, as inflorescências foram fixadas em solução de etanol/ácido acético glacial (3:1) por 24 h, transferidas para solução de etanol 70% e conservadas sob refrigeração. As mesmas foram submetidas à coloração com carmim acético e quantificado o número de micronúcleos em aproximadamente 300 tétrades para cada inflorescência, sendo confeccionadas lâminas com 10 inflorescências/semana para cada ponto estudado. As médias e desvios padrões foram: Dutra (2,24 ± 1,58), Estiva (2,07 ± 1,33), Parque Aeroporto (1,47 ± 1,05), Bom Conselho (1,42 ± 0,90) e Agronomia (0,82 ± 0,80). Apesar de Taubaté ser uma região em crescimento urbano, os dados revelaram que o ar da cidade apresentou boa qualidade. Manter esse equilíbrio é fundamental, requer esforços e aferição periódica, neste sentido, o biomonitoramento é um método rápido, barato e eficiente.
