Tumor-Suppressive miR-192-5p Has Prognostic Value in Pancreatic Ductal Adenocarcinoma

Abstract: Pancreatic ductal adenocarcinoma (PDAC) is characterized by fast tumor progression and diagnosis at advanced, inoperable stages. Previous studies could demonstrate an involvement of miR-192-5p in epigenetic regulation of visceral carcinomas. Due to contradictory results, however, the clinical utility of miR-192-5p in PDAC has yet to be determined. MiR-192-5p expression was analyzed by RT-qRT-PCR in human PDAC and benign tissue (n = 78), blood serum (n = 81) and serum exosomes (n = 74), as well as in PDAC cell … Show more

“…In the present context, expression of miR-181a-2-3p, miR-216b-5p and miR-214-5p were significantly different in PDAC tissue compared to CP tissue by both NGS and qRT-PCR but there was no significant difference in their serum levels between the two groups of patients. This observation is not unique to this study as many other reports show little or no concordance between tissue and circulatory levels of the same microRNA 24,40 . This indicates that due care should be given to determine the miRNA levels in both tissue and serum as miRNAs showing concordance would serve as better biomarkers.…”

“…Likewise, controversial results have been reported about the alteration of miR-192-5p in PDAC tissue specimens. While Flammang et al 40 and Botla et al 41 reported lower expression of miR-192-5p in PDAC tissue; others have reported increased expression in pancreatic cancer tissue 24,42,43 . In fact, ectopic expression of miR-192-5p associates with enhanced cell proliferation and migration, reduced apoptosis and promotes cell cycle progression in pancreatic cancer 43 .…”

“…29 miR-192-5p downregulation plays a tumor-inhibitor part and probably acts a diagnostic and prognostic biomarker in pancreatic ductal adenocarcinoma. 30 Curcumin inhibited cell viability, induced apoptosis, and inhibited the PI3K/Akt signaling pathway through the overexpression of miR-192-5p in human non-small-cell lung cancer. 31 miR-192 is upregulated in cervical cancer, precancerous tissue, and serum compared with the normal tissue, so miR-192 could be regarded as potential biomarker for early diagnosis of cervical cancer.…”

“…In addition to proteins also sEV-resident miRNAs were investigated as a potential diagnostic tool for the early detection of PDAC. Serum-derived sEVs from PDAC patients were reported to contain elevated levels of miR-192-5p, miR-19a-3p, and miR-19b-3p, when compared to healthy controls [ 195 , 196 ]. Additional studies described significantly more miR-10b, miR-17-5p and miR-21 in PDAC patient samples [ 197 , 198 , 199 , 200 ].…”
