BackgroundCrop landraces embody a source of beneficial genes potentially providing endurance to environmental stress and other agronomic qualities including yield. Our study included 88 snake melon accessions (Cucumis melo var. flexuosus) collected from 9 districts in the Palestinian West-Bank. These accessions represent four landraces of Palestinian snake melon: Green, and White Baladi, and Green, and White Sahouri.ResultsThis is the first report on successful application of genotyping by sequencing in snake melon. Nine thousand seven hundred fifty single-nucleotide polymorphism (SNP) and 7400 DArTseq genetic markers were employed to evaluate genetic biodiversity and population structure of Palestinian snake melon germplasm collection. Clustering based on neighbor-joining-analysis, principle coordinate and Bayesian model implemented in Structure showed that patterns of genetic diversity of snake melon landraces depends on their geographical source and unraveled the presence of two major local landraces (Sahouri, and Baladi) with accessions from each group clustering together. A significant correlation was observed between both types of markers in Mantel correlation test. A significant association between genetic and geographic matrices (P < 0.0001) was also detected. AMOVA indicated that majority of variation (90%) was due to the difference within accessions.ConclusionThe Palestinian landraces seem to have unique genes that may allow the enhancement of the global snake melon gene pool and developments of the plant production worldwide. Our subsequent objective is to detect genotypes with promising qualities and to conduct association mapping studies concentrating on Fusarium-wilt resistance, yield, and environmental stresses.Electronic supplementary materialThe online version of this article (10.1186/s12870-018-1475-2) contains supplementary material, which is available to authorized users.

The experiment have been done on the winter farming season 2020 in one of the farms that belongs to Faris company in Basrah governorate, the technique ISSR have been used to study the genetic distance for twenty one isolates of snake melon Cucumis melo var. flexuosus species. The variations between amplified samples have been revealed after running them on a gel of agarose which have been previously stained by ethidium bromide. Five primers which gave varied product on the agarose have been selected. Those five primers produced 713 bands, both primers UBC 813 and UBC 815 showed the higher numbers of bands reached to 177 while the primer UBC862 showed the least numbers of bands (100) and the bands which showed multi-variations showed (46) bands, and the results primers amplification unique bands their number reached to (14) and five of those bands belong to the primer UBC842 while the primer UBC862 produced three bands while the primer UBC807 did not produced any bands. While it has shown 100% polymorphisms with the primers 813, 815, 842 and 862, and the least polymorphism percentage have shown with the primer UBC807 reached 75%. According to the efficiency primers, the highest efficiency percentage shown with the primer UBC813 and 815 reached to 24.82% and the least percentage shown 14.02% by the primer UBC862. Cluster analysis showed the effect on the variance of the studied cultivars.

The present study describes the effects of medium, explant type and photoperiod on somatic embryogenesis of snake melon (Cucumis melo var. flexuosus). Two different types of explant (cotyledon and hypocotyls) obtained from 3 days old snake melon seedlings and culture conditions were investigated for somatic embryogenesis. Explants were cultured on Murashige and Skoog (1962) (MS) medium supplemented with 23 different combinations of auxins (2,4-dichlorophenoxyacetic acid, 2,4-D; α-naphthalene acetic acid, NAA) and cytokinin (6-benzylaminopurine, BAP; N6-[2isopentyl]adenine, 2iP) either alone or in combination with each other. Embryogenic calluses were obtained from cotyledon explant on MS media supplemented with NAA and 2,4-D alone and combinations with cytokinin. Our findings showed that NAA and 2,4-D did not act synergistically with BAP or 2iP. Callus formation was the same whether BAP or 2iP was added into the media or not. The highest embryo formation was achieved from cotyledon explant which was 20.00±7.94 somatic embryos per petri dish on medium supplemented with 4 mg/l 2,4-D and 0.5 mg/l BAP. The cotyledon and hypocotyls explants cultured on the media with 2 mg/l 2,4-D with 0.5 mg/l BAP showed embryogenic callus formation and number of somatic embryos were 17.33±5.51 and 16.33±3.06 per petri dish, respectively. Auxin was found critical for formation of somatic embryo and at the same time, the two tested cytokinins at any concentration acted synergistically with auxin.
