Unconventional secretion of Pichia pastoris Acb1 is dependent on GRASP protein, peroxisomal functions, and autophagosome formation

Abstract: Evidence is presented for an unconventional protein secretion pathway that is conserved from yeast to Dictyostelium discoideum in which Acb1 may be sequestered into autophagosomal vesicles, which then fuse (either directly or indirectly) with the plasma membrane (see also the companion paper from Duran et al. in this issue).

“…Extracellular conversion of a protein precursor into signaling peptides has also been observed in the distantly related dictyostelid Polysphondylium pallidum and the yeasts Pichia pastoris and Saccharomyces cerevisiae, as well as mammalian astrocytes (14,15,27,29). In Pichia, the SDF-2-like activity produced from the ACB1 precursor is required for quorum sensing to regulate sporulation (29).…”

“…18 More recently, in yeast strain Pichia pastoris, Spo14 has been implicated in unconventional exocytosis of Acb1, and the process involves the fusion of APs with the plasma membrane. 19 PLD catalyzes the removal of choline from PC, generating PA, that could then serve as a precursor for the production of diacylglycerol (DAG). When locally concentrated, both PA and DAG favor negative curvature in membranes, lowering the energy barrier to fusion.…”

“…Proper expression and secretion of a key antibacterial enzyme lysozyme likely requires functional autophagy. Recent evidence supports a critical role for autophagy in the processing of secreted proteins (29). The sensitivity of Paneth cells to high doses of Lys05 suggests that Paneth cell dysfunction could accompany other novel autophagy inhibitors that are currently under development.…”
