Uniform Widespread Nuclear Phosphorylation of Histone H2AX Is an Indicator of Lethal DNA Replication Stress

Abstract: Phosphorylated histone H2AX (γ-H2AX), a central player in the DNA damage response (DDR), serves as a biomarker of DNA double-strand break repair. Although DNA damage is generally visualized by the formation of γ-H2AX foci in injured nuclei, it is unclear whether the widespread uniform nuclear γ-H2AX (called pan-nuclear) pattern occurring upon intense replication stress (RS) is linked to DDR. Using a novel monoclonal antibody that binds exclusively to the phosphorylated C-terminus of H2AX, we demonstrate that H… Show more

“…Liver tumor sections were embedded in paraffin before being fixed in 8% buffered formalin solution for 12 h. Then the paraffin-embedded hepatic tumor tissues were analyzed for any expression of Bax, PUMA and p53 by immunohistochemistry as previously described [23]. Tumor tissue sections were incubated with primary anti-PUMA, anti-Bax from Cell Signaling Technology (Boston, MA, USA) and anti-P53 antibodies from Abcam (Boston, MA, USA).…”

“…57,58 In addition, a recent study has linked DNA-PKcs to replication stress-induced, pan-nuclear H2AX phosphorylation in S-phase cells. 59 To better understand the mechanism behind the enhanced, olaparib-induced H2AX phosphorylation in ATM-deficient cells, A549-CRISPR-control, A549-CRISPR-DNA-PKcs and A549-CRISPR-ATM cells were treated with 4 µM olaparib for 1-4 days as in Fig. 3, and analysed for γ-H2AX foci using immunofluorescence.…”

“…TOPK depletion in combination with HU treatment was also associated with an increase in cells displaying diffuse pan-nuclear γH2AX staining. Nuclear-wide formation of γH2AX occurs during catastrophic replication stress, and invariably precedes irreversible cell death [27].…”
