Venlafaxine Ingestion Is Associated With Rhabdomyolysis in Adults: A Case Series

Abstract: -Rhabdomyolysis has been reported after venlafaxine ingestion. We wished to characterize the prevalence of this adverse effect in a realistic clinical setting. Therefore, a retrospective casenote review was performed, including 235 patients admitted to the Royal Infirmary of Edinburgh due to venlafaxine overdose between January 2000 and June 2006. Seizures occurred in 8.9% of the study population. Patients who suffered seizures had ingested larger quantities of venlafaxine than those who did not develop seizur… Show more

“…It inhibits central serotonin and norepinephrine neuronal reuptake increasing the diminished levels of neurotransmitters like serotonin and norepinephrine in the synaptic cleft between neurons in the brain (Wilson et al, 2007). VLF is commercially available as immediate and controlled release tablets and capsules (Effexor, Effexor XR) but oral therapy exhibits number of drawbacks, such as slow onset of action, low bioavailability (40-45%), elimination half-life of 4-5 h along with side effects like tachycardia, increased blood pressure, fatigue, headache, dizziness, sexual dysfunction, dry mouth.…”

“…3 There is a much wider reported literature for venlafaxine ingestion in adults typically as episodes of deliberate self-harm, including polypharmacy overdoses. [4][5][6] Seizures, rhabdomyolysis and cardiotoxic effects are reported in adults. Dystonic reactions to venlafaxine have not been previously reported in adults or children.…”

“…It inhibits central serotonin and norepinephrine neuronal reuptake and has proven efficacy in the treatment of depression and anxiety disorders (8). The drug is effective in the treatment resistant depression and thought to be superior to selective serotonin reuptake inhibitors in preventing the recurrence of depression (9,10).…”

“…Dose dependant elevation of creatinine kinase has also been reported in the patients suffering from VH toxicity. High prevalence of acute muscle injury or rhabdomyolysis was observed with VH overdose too (8). Statistical data generated from the usage pattern of the drug had revealed a potential risk factor of fatal overdose suggesting the need for cautioning patients with suicidal ideation (18).…”
