Xylo-oligosaccharides from sugarcane show prebiotic potential in a dynamic computer-controlled in vitro model of the adult human large intestine

Abstract: The aim of the study was to investigate the prebiotic potential of xylo-oligosaccharides (XOS) from sugarcane in a validated, dynamic, computer-controlled in vitro model of the colon (TIM-2) simulating human adults. In two sets of experiments, each with a different microbiota, 3 different doses of XOS were tested at 1.0 g/day, 1.5 g/day and 3.0 g/day. The in vitro model was run for 72 h, and at the start and subsequently every 24 h samples were taken and analysed for short-chain fatty acids (SCFA) and gut micr… Show more

“…DNA isolation and sequencing of barcoded amplicons of the V3-V4 region of the 16S rRNA gene were essentially performed as described before [25] according to established protocols provided by Illumina (Illumina, Eindhoven, the Netherlands). In brief, barcoded amplicons from the V3-V4 region of 16S rRNA genes were generated using a 2-step PCR.…”

“…The stabilization period is essential in dynamic models, and it is performed before starting the experiment. The followed protocol depends on the system characteristics [62,73,74]. Furthermore, achieving the physiological conditions of the environment is a key aspect for reaching the proper simulated microbiota in each colonic region.…”

“…Furthermore, achieving the physiological conditions of the environment is a key aspect for reaching the proper simulated microbiota in each colonic region. Consequently, these models work at 37 • C, maintain anaerobic conditions (e.g., by periodically flushing nitrogen), control the pH by automatically adding NaOH or HCl to each vessel, and mixing the compartment contents (e.g., by magnetic stirring or peristaltic movements) [73,75]. Finally, colonic variables such as pH, retention time, and volume not only depend on the utilized model but also on the aim or purpose of the study.…”

“…In this context, the use of diet as a basis for modifying the microbiota has re-emerged in recent years, validating the ancient concepts of the relevant role of nutrition in health (Requena et al, 2018). Therefore, the interest for probiotics or novel sources of prebiotic compounds is increasing all over the world (Ewaschuk and Dieleman, 2006;Gareau et al, 2010;Alagón Fernández del Campo et al, 2019;Venema et al, 2020).…”
